酸甘化阴、辛甘化阳是“五味合化”理论的一个应用典范。即酸甘味药合用，能滋阴养血；辛甘味药配伍，可温运阳气。其临床应用非常广泛，但是缺乏实验研究。当归芍药散在临床及实验研究中发现对于阿尔茨海默病（Alzheimer’s Disease，AD）有良好的改善作用，从处方组成来看，恰好蕴藏着上述药性配伍理论。研究发现AD的发病机制跟自噬高度相关，而小胶质细胞内cAMP/cGMP的浓度水平是触发Ras/cAMP-dependent PKA通路导致自噬的主要原因，以往研究认为cAMP和cGMP相互拮抗，相互依存，这一对环核苷酸可能是中医阴阳学说的物质基础。因此本课题运用拆方研究方法，利用分子生物学技术考察各组对人源型小胶质细胞AD模型的cAMP/cGMP的浓度水平与自噬通路和神经元功能的影响，揭示当归芍药散通过酸甘化阴、辛甘化阳配伍防治AD的作用机制，丰富中药药性理论的内涵。

Combination sour and sweet herbs nourishing yin and combination acrid and sweet herbs activating yang, which was a good example application of TCM theory of five tastes of medicinal herb combined with each other. That was, .the combination of sour and sweet medicines could nourish Yin and blood, and the compatibility of acrid and sweet medicines could warm Yang Qi. This theory in clinical application was very extensive, but lack of experimental research data. In previous studies, it was found that Danggui Shaoyao powder had a good effect on the improvement of Alzheimer's disease (AD). In terms of composition, it contained the property theory above-mentioned. Furthermore, it was found that the pathogenesis of AD was highly correlated with autophagy. The concentration of cAMP/cGMP in microglia was the main cause of autophagy triggered by Ras/cAMP-dependent PKA. Previous studies had also suggested that cAMP and cGMP were antagonistic and interdependent, and this pair of cyclic nucleotides may be the material basis of Yin-Yang theory of TCM. Therefore, by using the method of decomposition and molecular biology, we are going to investigate the concentration of cAMP/cGMP in human microglia AD model, the effect of autophagy pathway and neuron function, in order to verify the property theory of combination sour and sweet herbs nourishing yin and combination acrid and sweet herbs activating yang. This study will reveal the prescription regularity of traditional Chinese medicine and the mechanism of preventing and treating AD, and enrich the compatibility theory of traditional Chinese medicine.
