孤独症是一种遗传度较高的神经发育障碍。社会交往受损是其主要核心症状之一。研究提示神经环路的精细结构和脑区间的信息传递在社交行为中发挥重要作用。既往报道孤独症患者存在社交辨识记忆障碍和齿状回(DG)发育异常表型,且本项目前期研究发现孤独症易感基因缺失导致的DG发育异常可能参与小鼠模型的社交辨识缺陷,但其内在因果关系及神经环路机制尚未完全明确。本项目拟通过活动特异性激活标记、光纤记录等方法明确参与社交辨识行为的DG相关神经环路和神经元类型;利用免疫荧光染色、电生理记录等方法系统评价神经环路的突触连接、结构和功能;进一步通过化学遗传和光遗传学操控特异神经环路的活动性,观察对小鼠社交辨识行为的影响。本项目预期结果将初步揭示DG发育异常导致的相关神经环路结构和功能缺陷在孤独症社交辨识障碍中的病理基础,并有助于理解社交辨识记忆的神经环路编码和信息传递机制,为寻找孤独症等社交缺陷疾病的治疗靶点提供线索。
英文摘要
Autism is a neurodevelopmental disorder with high heritability. Impairment of social interaction is one of the core symptoms of autism. Researches suggested that the fine structure of neural circuits and information transmission among brain areas play important roles in social behaviors. Previous studies reported impaired social recognition and abnormal development of dentate gyrus (DG) in autistic patients. In addition, our preliminary work showed that the abnormality of DG development might be involved in social recognition deficit of autistic susceptibility gene knockout mice models. However, the causal relationship and underlying neural circuitry mechanism remain unclear. In this study, the roles of DG related neural circuits in social recognition behavior will be investigated using activity-dependent labeling and optical fiber recording. The synaptic connection, structure and function in neural circuits will be systematically assessed using immunofluorescence staining and electrophysiological recording. The effects of neural circuits’ activation/inhibition on social recognition behavior will be explored using chemogenetics and optogenetics. This study will reveal pathological mechanism of social recognition deficit including the structural and functional defects of neural circuits due to the abnormal DG development in autism. In addition, it will be helpful to understand the neural circuitry mechanism and information transmission of social recognition memory, and provide clues for investigating the therapeutic targets of social defect disorders including autism.
孤独症谱系障碍(ASD)是一组高度遗传的神经发育障碍。因发病机制未明,故而缺乏有效的干预手段。既往神经病理学和影像学研究报道患者存在海马齿状回(DG)的发育异常,包括体积减小和颗粒细胞排列紊乱、异位等表型。然而,DG发育不良的潜在遗传因素和DG发育不良导致相关神经环路信号传递受损在孤独症社交障碍中的机制却鲜有研究。本项目发现易感基因Auts2在出生后早期DG发育过程中与Ctip2结合,通过招募NURD转录抑制复合物来抑制Ptgds转录。DG的分裂后颗粒细胞中出现Ptgds的异位高表达,并通过非细胞自主方式抑制神经干细胞(NSC)/中间前体细胞(IPC)从软脑膜下区(SPZ)向颗粒下区(SGZ)的迁移,导致出生后早期DG体积减小和SGZ区形成障碍。此外,还观察到孤独症易感基因Trio的缺失导致小鼠出生后DG区体积减小且DG上回颗粒细胞排列呈锯齿状排列。主要原因为前体细胞(IPCs)的增殖和迁移过程受阻,IPCs的迁移流和分裂后颗粒细胞的迁移流发生异常交汇,使得局部IPCs发生聚集,细胞迁移和定位出现异常。空间转录组测序发现,Trio在DG不同发育阶段的颗粒细胞中的表达模式和功能不同:在分裂后颗粒细胞中与调控神经元迁移的信号通路最为相关;而在IPCs中参与细胞连接和信号转导等功能。这些发现从神经发育角度解释了临床研究发现DG发育不良参与ASD发病的潜在机制。在此基础上,本项目还发现DG发育不良影响下丘脑乳头体上核(SuM)-DG-CA3神经环路信号传递,导致SuM区对海马的神经调控出现异常并参与社交辨识行为障碍。通过激活下丘脑-海马神经环路,或通过多种策略促进DG区神经元的新生,重塑DG局部细胞构筑,可以通过提升DG-CA3环路的突触传递,部分挽救小鼠的社交辨识缺陷,为理解ASD社交障碍的致病机制和发现潜在干预策略提供新的依据。
