早期诊断对于胰腺导管腺癌(PDAC)的治疗和预后具有重要意义,因此发展有效的新型分子影像学手段成为临床迫切需求。Kras基因介导的信号通路及谷氨酰胺代谢重编程在PDAC的发生、发展和耐药机制中发挥着关键作用。本项目拟以PDAC肿瘤的代谢机制和诊治为向导,构建放射性核素标记的谷氨酰胺代谢分子探针,发展PDAC体内检测新方法,并探索肿瘤谷氨酰胺代谢与Kras基因调控相关性。本项目首次探究了多参数PET/MRI谷氨酰胺代谢显像在PDAC诊断中的应用价值和优势,以期为PDAC患者筛选、治疗方案优化及预后评估提供新策略,有效协助临床对肿瘤患者进行分层及个体化诊疗。
英文摘要
Early diagnosis has great significance for the efficient treatment and better prognosis of pancreatic ductal adenocarcinoma. Therefore, the development of effective new molecular imaging methods has become an urgent clinical need. The Kras gene-mediated signaling pathway and glutamine metabolism reprogramming play a key role in the occurrence, development, and resistance of PDAC. Guided by the metabolic mechanism and diagnosis and treatment of PDAC tumor, this project intends to construct a radionuclide-labeled molecular probe for glutamine metabolism, develop a new method for in vivo detection of PDAC, and explore the relationship between tumor glutamine metabolism and Kras gene regulation. This project explores the value and advantages of multi-parameter PET/MRI metabolic imaging of glutamine in the diagnosis of PDAC for the first time, in order to provide a new strategy for the screening, treatment program optimization and prognosis evaluation of PDAC patients, so that it can effectively assist the clinical stratification and individualized diagnosis and treatment of tumor patients.
