我们研究发现肝癌中WISP1mRNA高表达而蛋白低表达，其mRNA和蛋白表达显著不一致[2018,hepatology]，这种现象可能与mRNA翻译异常有关。然而，WISP1mRNA翻译异常是否与其不一致性有关仍不清楚。研究显示LncRNA可调控mRNA翻译。前期RNA-Seq发现: 肝癌中LncRNA-WISP1-AS（WISP1-AS）高表达；下调WISP1-AS表达导致WISP1蛋白表达增加，而mRNA表达不变，其mRNA和蛋白不一致程度下降，且抑制肝癌细胞增殖；下调WISP1-AS表达同时干扰WISP1表达回复了肝癌细胞增殖；生物软件预测WISP1-AS可与WISP1mRNA结合。据此推测：WISP1-AS破坏WISP1mRNA和蛋白一致性促进肝癌细胞增殖。为验证推测，本课题拟明确WISP1-AS破坏WISP1mRNA和蛋白一致性机制，为肝癌防治提供新的理论依据。

We have found that WISP1 mRNA is highly expressed and WISP1 protein low expressed in hepatocellular carcinoma. The WISP1 WISP1 mRNA and protein expression is significantly discordant. [2018, hepatology]. This phenomenon may be related to abnormal translation of WISP1 mRNA. However, it is not clear whether the abnormal translation of WISP1 is related to its discordance. Studies have shown that LncRNA can regulate the translation of mRNA. Previous findings in RNA-Seq: high expression of LncRNA-WISP1-AS (WISP1-AS) in hepatocellular carcinoma; Down-regulation of WISP1-AS resulted in increased expression of WISP1 protein , while the expression of WISP1-AS remained unchanged, the degree of discordance between WISP1mRNA and protein decreased, and inhibited the proliferation of hepatocellular carcinoma cells; Interfering WISP1 expression also to restored the proliferation of hepatocellular carcinoma cells with down-regulation of WISP1-AS expression; Biosoftware predicted that WISP1-AS could bind WISP1-AS. It is speculated that WISP1-AS can destroy the accordance of WISP1 mRNA and protein and promote the proliferation of hepatocellular carcinoma cells. In order to verify the hypothesis, we intend to clarify the mechanism of WISP1-AS destroying the accordance of WISP1 mRNA and protein, which provides a new theoretical basis for the prevention and treatment of hepatocellular carcinoma.
