卵巢癌早期症状不明显和临床上早期预警标记的缺乏是导致卵巢癌患者死亡率高的重要原因之一。探索发现敏感的、非侵入性的生物标记物来提高卵巢癌的早期检测准确率一直是卵巢癌研究的热点和难点问题。miRNA的癌相关性和组织特异性及其在血清中的稳定性和可检测性,使血清中的miRNA成为卵巢癌早期预警标记研究的新希望。然而,血清中信号来源复杂性、风险预测模型的不稳定性和标记特异性评估缺乏,导致了目前在血清中发现的卵巢癌miRNA风险标记存在重复性差和临床转化率低的现象。本项目拟在前期分析外周全血中疾病相关信号来源及免疫损伤标记识别的研究基础上,进一步探索血清中卵巢癌相关的miRNA风险标记。主要研究内容包括:(1)基于miRNA间相对表达大小关系构建非肿瘤血清样本中miRNA表达模式参考系;(2)血清中卵巢癌特异的miRNA风险预警标记识别;(3)血清中卵巢癌miRNA风险标记的来源及机理分析。

The unclear early clinical symptoms and lack of early diagnostic biomarkers is one of the important reasons for the high mortality of patients with ovarian cancer. Detecting sensitive and non-invasive biomarkers to improve the accuracy of early diagnostic of ovarian cancer has been a hot and difficult issue in ovarian cancer research. The characteristics of association with cancer, tissue specificity and their stability and detectability in serum make microRNAs (miRNAs) become a new hope for detecting early diagnostic biomarkers of ovarian cancer. However, the complexity of signal sources, the instability of risk prediction models, and the lack of biomarker specific assessment have led to the poor reproducibility and low clinical conversion rate of reported ovarian cancer miRNA biomarkers. This project intends to further explore the ovarian cancer associated miRNA biomarkers in serum based on the experience about analyzing the source of disease-associated biomarkers and identifying immune damage markers in peripheral blood. The main research contents include: (1) Constructing a reference frame of miRNA expression patterns in non-cancer serum samples based on the rank of relative expression between miRNAs. (2) Identification ovarian cancer-specific miRNA biomarkers in serum. (3) Analyzing the source and mechanism of ovarian cancer associated miRNA biomarkers detected from serum.
