With the increasing antibiotic resistance of pathogenic strain Staphylococcus aureus, the development of new control methods is imminent. Bacterial RNA polymerase is a super-complex consisting of five subunits (2α, β, β' and ω), the sequence and three-dimensional structure of which are highly conserved in most bacteria, but different from human RNA polymerase, therefore bacterial RNA polymerase is an important drug target for new anti-infective drug development. Staphylococcus aureus RNA polymerase has a unique δ subunit, which has been proved to directly participate in the expression of virulence factors genes. The deletion of the δ subunit can significantly reduce the cell adhesion, anti-immune cell clearance and host cell diffusion ability of Staphylococcus aureus strains. Targeting Staphylococcus aureus RNA polymerase with δ subunit may be a novel method to control antibiotic resistant strains. However, the structure of Staphylococcus aureus δ subunit, the structure of Staphylococcus aureus RNA polymerase complexed with δ subunit, and the transcriptional characteristics and regulatory mechanisms of the complexes have not yet been clarified and elucidated.. We have previously expressed, purified and reconstituted the Staphylococcus aureus RNA polymerase holoenzyme containing δ subunit in vitro successfully. In this project, we plan to solve the three-dimensional structure of transcription activation, transcription pause and transcription termination complexes of Staphylococcus aureus, and further characterize the biological functions of the δ subunit in transcriptional initiation, elongation and termination of Staphylococcus aureus in vitro and in vivo. The project will provide the experimental basis for the design of anti-infective drugs targeting this molecular machine by solving this unique transcriptional molecular machine structure of important pathogen S. aureus.
进入 21 世纪以来,由于传统抗病原菌感染药物被广泛的应用于临床和畜牧业,大量的抗菌药物残留于环境中,造成耐药菌在动物—环境—人类中的交互传播,导致病原菌耐药性在全球范围内的快速上升,病原菌耐药性的快速上升正严重危胁着人类的生存与健康。因此,急需鉴定病原菌新药物靶标并解析其作用机制,以便更好的研发新型抗耐药菌感染的药物。. 本研究第一部分,运用结构生物学、定点突变和生化方法揭示了金黄色葡萄球菌δ亚基如何与RNA聚合酶核心酶和DNA相互作用,解析了δ亚基调控RNA聚合酶活力和转录的分子机制,加深了对delta亚基在金葡菌转录调控过程中发挥潜在作用的理解,为新型抗耐药金黄色葡萄球菌感染药物研发提供了新的药物靶标。. 本研究第二部分,采用低温冷冻电镜技术解析了病原菌多效性转录调控蛋白Rob依赖型转录激活复合物(Rob-dependent transcription activation complex,Rob-TAC)的结构,并通过生化实验鉴定并验证了Rob与RNAP、DNA之间的关键相互作用,阐明了Rob CTD在转录激活过程中发挥的关键别构调节作用,揭示了多效性转录调控蛋白Rob激活转录的分子机制,为理解病原菌AraC家族蛋白激活感染、致病与耐药基因转录的分子机制奠定了重要基础,同时也为新型抗耐药菌药物的研发提供新的靶标与研究思路。. 本研究第三部分,利用生物化学、微生物遗传学、结构生物学揭示了GlnR的转录调控活力不受双组份系统应答调控蛋白的天冬氨酸磷酸化修饰调节;解析了结核分枝杆菌GlnR的N端信号接收结构域(REC)、C端DNA结合结构域(DBD)与cis-element、及其RNA聚合酶的复合物结构,揭示了GlnR蛋白采用同源四聚体而非二聚体稳定转录激活复合物这一全新的微生物基因转录调控模式,阐明了GlnR以四聚体形式调控靶标基因转录的生理意义,即:可以更快、更精确的响应外界营养匮乏等应激信号和调控更多的下游靶标基因,这些结果为抗耐药结核菌感染药物的研发提供了新的思路和靶标。
