1,3-Butadiene (BD) is a kind of important industrial material, that is widely used in the production of resins and plastics such as butadiene rubber and polychloroprene. The toxicological effects of butadiene have been given more attention. In order to get details about cloning efficiency, mutation frequency, the spectra, mechanism and repair of HPRT gene mutation induced by butadiene, blood lymphocytes of BD-exposed workers and control subjects were examined by methods of cell clone culture, two-way screening count, multiplex PCR amplification, electrophoresis and transgenic techniques. The number for BD-exposed workers and control subjects was 74 and 157, respectively. It was discovered that mutation frequency of BD-exposed workers was higher than control subjects', but not significantly (P>0.05). The percentage of deletion mutation in BD-exposed subjects was bigger than that in control subjects significantly (P<0.05), in particular multiplex exons often deleted at the same time. Allied results were got in vitro tests, too. It suggested that BD was liable to cause bigger changes in genetic structure, which showed health of BD-exposed workers could not be protected in present country healthy standard (100mg/m3). This is the first time that heredity affect of BD-exposed workers were investigated in system in our country. The research could give some help to afresh country BD healthy standard, to build the international database of HPRT gene mutation and to study mechanism of genetic disease with HPRT lack.
