CaV1.2 plays an indispensable and important role in the regulation of dental pulp stem cell (DPSC) odontogenic and neurogenic differentiation. Previous studies of CaV1.2 pathway on DPSC differentiation are mostly phenomenon studies based on the negative regulation of calcium ion channels. Although the necessity of CaV1.2 in DPSC differentiation has been confirmed, the molecular mechanism of it has not yet been clarified. Therefore, it has limited the clinical development of new drugs for dental pulp repair. This project is based on the dental pulp injury model, focusing on activation regulation, to study how CaV1.2 regulates DPSC odontogenic and neurogenic differentiation processes. By analyzing the three-dimensional structure of CaV1.2 protein, we explore the combining mode between small molecular agonist and CaV1.2 protein to obtain docking information, aiming to establish a working framework which is guiding the design and development of small molecular drugs. Finally, we will use injury model of dental pulp to verify the potential of new drugs in repair. The implementation of this project will clarify the molecular mechanism of CaV1.2 to repair injury dental pulp, guide the development of new drugs, promote clinical transformation, and provide a new way to therapy damaged dental pulp in the future.
牙髓干细胞(Dental pulp stem cells, DPSCs)因其快速增殖和多向分化能力,在牙齿修复和牙髓再生领域展现出重要应用潜力。本研究旨在探讨L型钙离子通道(L-type calcium channel, LTCC)激动剂BayK8644对人牙髓干细胞(hDPSCs)成牙向分化的影响及其潜在机制。首先,通过体外实验优化BayK8644的适宜浓度,发现其在1 μM浓度下显著促进hDPSCs的碱性磷酸酶(ALP)表达、矿化结节形成及成牙相关基因(ALP、DSPP和OCN)和蛋白的表达。随后,通过半原位牙髓再生动物模型验证BayK8644的体内效果,结果显示其可显著促进矿化物形成并上调成牙本质样细胞的DSPP表达水平。机制研究表明,BayK8644通过激活MAPK/ERK信号通路促进ERK磷酸化,从而调控hDPSCs的成牙向分化;ERK抑制剂u0126的使用可显著抑制BayK8644的促分化作用。综上所述,本研究揭示了BayK8644促进hDPSCs成牙向分化的作用及其机制,为LTCC激动剂在牙髓再生中的应用提供了新的理论依据和实践支持。
