不明原因复发性流产(URSA)的免疫调节机制尚不明确。前期我们发现在复发性流产蜕膜巨噬细胞中,lncRNA-AOC4P的表达显著上调,而miR-410-5P表达明显下调,且lncRNA-AOC4P可显著上调GTPBP10,而miR-410-5P对GTPBP10具有负反馈调节作用。lncRNA-AOC4P、miR-410-5P和GTPBP10与慢性炎症、组织重建及免疫细胞功能异常有关。推测lncRNA-AOC4P可能通过改变miR-410-5P水平,从而靶向促进GTPBP10介导的URSA诱发。本研究拟通过RIP、双荧光素酶报告系统等分析lncRNA-AOC4P、miR-410-5P及GTPBP10之间的调节关系,运用FCM及细胞共培养等实验技术分析免疫细胞的分化以及细胞间的相互作用,为URSA找到可能的免疫调节机制。

The immunomodulating mechanism of unexplained recurrent spontaneous abortion (URSA) remains unclear. Previously, we found that the expression of lncRNA-AOC4P was significantly increased, the expression of microRNA-410-5P was significantly decreased, and the expression of GTPBP10 was significantly increased in decidual macrophages of URSA. lncRNA-AOC4P could significantly up regulate GTPBP10, while microRNA-410-5P could down regulate GTPBP10. LncRNA-AOC4P, microRNA-410-5P and GTPBP10 are associated with chronic inflammation, tissue remodeling and abnormal immune function of cells. It is speculated that lncRNA-AOC4P may be involved in the immunoregulatory of URSA by targeting GTPBP10 through the mediation of microRNA-410-5P. In this study, the regulation of lncRNA-AOC4P, microRNA-410-5P and GTPBP10 will be analyzed by RIP and double luciferase reporting system,and the differentiation of immune cells and the interaction between cells will be analyzed by FCM and co-culture experiments, so as to find a possible immunoregulatory mechanism for URSA.
