白术为临床常用大宗品种,生白术和泔制白术治疗溃疡性结肠炎(UC)已为临床验证,课题前期已明确其泔制工艺具有科学合理性,炮制后体外化学成分变化明显,具有补脾作用。基于此,本课题采用LC-MS/MS指纹轮廓谱和MALDI-MS质谱成像分析方法,全面动态表征白术炮制前后体内化学成分变化规律,识别白术、泔制白术的关键入体活性成分。通过DSS诱导复制UC小鼠模型,动态观察正常及疾病状态下UC的肠黏膜屏障结构、功能、损伤机制,运用代谢组学、蛋白质组学、质谱成像、16S rRNA基因测序、Western-Blot分析等手段,探讨白术和泔制白术药效作用,阐明泔制白术通过调节机械屏障claudin-2蛋白、化学屏障黏蛋白MUC、免疫屏障免疫球蛋白SIgA表达及改善生物屏障菌群,同时调控相关信号通路,综合阐释白术泔制增强健脾止泻效应的物质基础及作用机制,为指导临床治疗提供理论依据。

Atractylodis Macrocephalae is a common clinical variety.The treatment of ulcerative colitis ( UC ) by raw Atractylodes macrocephala Koidz and swill Atractylodes macrocephala Koidz has been clinically verified. The swill processing technology has been confirmed to be scientific and reasonable in the early stage of the subject. The chemical composition changes in vitro after processing are obvious, and it has the effect of tonifying the spleen. Based on this, LC-MS / MS fingerprint profile and MALDI-MS mass spectrometry imaging analysis method were used to comprehensively and dynamically characterize the changes of chemical components in Atractylodes macrocephala before and after processing, and identify the key active ingredients of Atractylodes macrocephala and Atractylodes macrocephala swill. The UC mouse model was induced by DSS to dynamically observe the intestinal mucosal barrier structure, function and damage mechanism of UC under normal and disease conditions. Metabonomics, proteomics, mass spectrometry imaging, 16S rRNA gene sequencing, and Western Blot analysis were used to explore the efficacy of Atractylodes macrocephala Koidz and Atractylodes macrocephala Koidz made from swill. It was clarified that Atractylodes macrocephala Koidz made from swill could regulate the expression of claudin-2 protein,MUC, and SIgA to improve the biological barrier flora, and regulate related signaling pathways, comprehensively explain the material basis and mechanism enhancing the effect of invigorating spleen and relieving diarrhea, and provide a theoretical basis for guiding clinical treatment.
本研究以临床常用大宗中药白术为研究对象,运用我国独有的中药炮制制药技术炮制“米泔水漂制白术”,根据自古以来临床医家“生熟异用”经验,研究泔制白术炮制增效作用机制。首先,将泔制白术动态划分5个阶段,采用HS-GC-MS及多元统计分析方法,鉴定49个泔制白术挥发性成分,阐释苍术酮、大根香叶烯B等关键成分在不同炮制阶段变化规律;采用UPLC-Q-TOF-MS和多元统计分析方法,鉴定55个泔制白术非挥发性成分,并阐释白术内酯Ⅰ、白术内酯Ⅱ、白术内酯Ⅲ等关键成分变化规律,进一步阐明泔制白术炮制工艺科学性。其次,将生白术及其泔制、麸炒、土炒、炒焦共5种白术不同饮片进行成分比较,基于HS-GC-MS鉴定出73种挥发性成分、并比较其不同饮片中差异情况;基于UPLC-Q-TOF-MS鉴定出57种化学成分,及其在不同饮片分组中变化规律,为阐明泔制白术药效物质奠定基础。再次,开展泔制白术药效作用研究,明确泔制白术有15个共有入血成分,并通过MAIDI-TOF/MS确认白术内酯III在结肠不同时间点的空间分布,可能是其发挥药效作用的关键物质基础。米泔水漂白术增强健脾止泻作用具体表现为,可改善脾虚泄泻小鼠的精神状态、被毛情况、大便形状等,体质量也在缓慢增加。生白术组和泔制白术组小鼠血清TNF-α的水平均显著降低(P<0.05),但泔制组的降低程度较生品组稍显著;与模型组相比,生品组和泔制组小鼠血清中VIP的水平均降低,但泔制组的降低程度有显著性(P<0.05)。泔制白术可改善DSS诱导的小鼠结肠炎症状,降低结肠组织中IL-1β和IL-6水平、提高IL-10表达水平。运用代谢组学与蛋白组学技术,富集了甘油磷脂代谢等信号通路。采用Illumina高通量测序结果表明,泔制白术对肠道菌群的Chao1、Ace、Shannon、Simpson、PD_whole_tree指数具有显著性差异,各给药组含肠道菌群数目大小为:漂白术组>麸炒白术组>焦白术组≈生白术组>土炒白术组。Lactobacillales是泔制白术的优势菌群,其酶活性特征可促使β-低聚半乳糖及GOS的产生,益于肠道菌群健康。
