基于“络脉”理论探讨通心络胶囊对糖尿病心肌病TGF-β1-p38MAPK-CREB信号通路的影响

糖尿病心肌病（DCM）是糖尿病的慢性并发症，其中心肌纤维化是糖尿病心肌病特征性病理表现。现有研究表明，糖尿病状态下心脏TGF-β1-p38MAPK-CREB信号通路激活与心肌纤维化的发生、发展有着密切关系。我们前期研究表明中药通心络胶囊是在成纤维细胞增殖水平上抑制了糖尿病心肌的纤维化。基于此，项目从通心络胶囊抑制糖尿病心肌病心肌间质纤维化的作用与机制研究入手，通过采用高脂饮食及小剂量链脈佐菌素(STZ)腹腔注射来诱导糖尿病心肌病(DCM)模型,并采用不同剂量的通心络进行干预，运用HE染色、Masson染色、免疫组化、免疫印迹、酶联免疫吸附等技术检测通心络处理能否改善其心肌间质纤维化、心脏功能，并探讨通心络胶囊对心肌组织内TGF-β1-p38MAPK-CREB通路相关因子的表达水平的影响，为中药防治糖尿病心肌纤维化寻找新的靶点。

Diabetic cardiomyopathy is a chronic complication of diabetes, and myocardial fibrosis is the characteristic pathological manifestation of diabetic cardiomyopathy. Existing studies have shown that the activation of cardiac TGF-beta 1-p38MAPK-CREB signaling pathway is closely related to the occurrence and development of myocardial fibrosis in diabetic patients. Our previous studies showed that Tongxinluo capsule inhibited diabetic myocardial fibrosis at the level of fibroblast proliferation. Based on this, the project started with the study of the effect and mechanism of Tongxinluo Capsule on inhibiting myocardial interstitial fibrosis in diabetic cardiomyopathy. The model of diabetic cardiomyopathy was induced by high-fat diet and intraperitoneal injection of low-dose streptozotocin. Different doses of Tongxinluo were used for intervention. HE staining, Masson staining, immunohistochemistry, immunoblotting and enzyme-linked immunosorbent assay were used. To explore the effect of Tongxinluo Capsule on the expression of TGF-beta 1-p38MAPK-CREB pathway related factors in myocardial tissue, and to find a new target for the prevention and treatment of diabetic myocardial fibrosis by traditional Chinese medicine.
