本课题拟从网络药理学多靶点的研究策略与中医药整体观、辨证论治原则相结合的角度运用现代生物技术对国家级名老中医王力宁教授治疗儿童CVA的验方麻杏二陈颗粒进行挖掘研究。通过网络药理学技术利用TCMSP平台查找麻杏二陈颗粒中各味中药含有的化学成分;检索哮喘相关的基因;用Cytoscape软件构建"药物-成分-靶标-疾病"网络图,确定网络关键节点;通过GO功能富集分祈和通路富集分析,对相关的受体信号通路构建匹配基因的相互作用网络,筛选关键的信号通路。并通过OVA致敏幼龄小鼠构建CVA动物模型,观察麻杏二陈颗粒对小鼠的肺部炎症及MAPK信号通路的影响,以期与网络药理学分析结果相互验证,有助于发现麻杏二陈颗粒的组效关系及其方剂配伍规律,探讨麻杏二陈颗粒治疗儿童CVA的作用机制,为进一步挖掘整理传承名老中医学术经验提供科学实验依据。研究成果将造福于广大CVA患儿,促进小儿身心健康与广西民族区域经济发展。
英文摘要
With the multi-target strategy of network pharmacology, based on the holistic view of traditional Chinese medicine and the principles of treatment combining syndrome differentiation, applying the contemporary biotechnology, the Project study the Maxing Erchen Granule(Compound of Chinese medicine,by Prof. Lining Wang)treating children CVA . First ,by the application of network pharmacology, TCMSP was applied to study the chemical components in the Chinese materia medica from Maxing Erchen Decotion; Secondly search asthma related genes in genetic database; Thirdly build up the Medicine-Composition-Target-Disease network with Cytoscabe software and confirm the network keypoints face to face. And then through the enrichment analysis of the GO function enrichment analysis and pathway enrichment analysis, the key genes were found by constructing a gene-matching network with receptor signaling pathway. In the next place, the CVA animal model was built by using OVA asthma mice. Through CVA model, the Maxing Erchen Granules’ effect on mice lung inflammation and its relevance to MAPK signaling pathway were inspected. We expect that mutual authentication between this result and network pharmacological study will benefit to discover the formula-activity relationship and formula composition rules of Maxing Erchen Granules, so as to explore the mechanism of children CVA treatment effect of Maxing Erchen Granules, and to provide scientific experimental evidences for further excavating and sorting out the academic experience of Reputed Master of Chinese Medicine. The results will benefit to the children with CVA and promote the development of children's health and local economy in ethnic region of Guangxi.
