During DNA replication, the genome is replicatied to generate daughter DNAs through defined replication timing program,and the histones also need to be re-assembled to the two daughter DNAs to form nascent chromatin. The inheritance of epigenetic information on the nascent chromatin is critical for the maintenance of genome stability and cell fate. The histone variant H2A.Z can modulate H4K20me2 levels to regulate the licencing and activation of early replication origins, and further to regulate the replication timing program.In this study, we plan to investigate how H2A.Z and H4K20me2 are distruibuted on daughter chromatin after DNA replication, and how H2A.Z and H4K20me2 are restored before next S phase.This project will offer new insights into the inheritance of epigenetic information after DNA replication, and into the inheritance of DNA replication origins and replication timing regulated by epigenetic information.
