血小板T细胞活化抗原1（PTA1）是一种新的与T细胞分化和血小板活化凝集等功能相关的白赴只乖颐亲罱⑾制湓谛∈笥肴薖TA1有相似的功能。本项目拟首先克隆小鼠PTA1cDNA序列，确定其基因的染色体定位，然后应用基因探针对PGA1在小鼠组织和器官中表达与植嫉墓媛山醒芯浚狙芯拷狿TA1转基因和基因敲除等进一步的研究打下基础。

Platelet and T cell activation antigen 1 (PTA1)/CD226 is a novel leukocyte differentiation antigen involving in the differentiation of T cells and aggregation of platelets. The cDNA of CD226 was cloned in 1996 by DNAX Institute in United States and in 1997 by our department cooperating with Newcastle University in Australia, respectively. The genes of gibbon and monkey CD226 were also cloned in 1997 in our department .Mouse is an ideal model in immunological research, especially in the functional study of a novel molecule and its relationship with some diseases by transgenic mouse and gene knock. We first cloned the cDNA of mouse PTA1 (mPTA1) and its three isoforms. Based on the expression of the mPTA1 fusion protein using prokaryotic and eukaryotic expression system, the monoclonal and polyclonal antibodies were prepared which were used for identification of mPTA1 distribution. The results showed that the mPTA1 is mainly expressed on CD8+ signal positive cells either in mouse thymocytes or perpheral lymph organs detected by flow cytometry. These results will help us to further study the in vivo function of mPTA1 by transgenic mouse and gene konckout.
