采后香蕉果实在贮运过程中极易发生冷害,导致果实品质劣变,失去商品价值。活性氧(ROS)与WRKY转录因子在植物逆境胁迫响应中发挥重要作用,而WRKY转录因子是否参与调控采后香蕉果实冷胁迫响应中的ROS代谢及其具体机制如何,尚不清楚。本项目在明确采后香蕉果实在冷胁迫响应过程中ROS合成与清除的生理变化规律的基础上;进一步基于转录组数据获得响应冷胁迫的WRKY转录因子,以及参与ROS代谢的下游结构基因;通过DNA-蛋白质互作筛选、鉴定调控ROS代谢相关结构基因的核心WRKY;采用蛋白质-蛋白质互作挖掘核心WRKY的互作蛋白,并分析互作蛋白对WRKY转录调控效应的影响。本项目将系统阐明采后香蕉果实冷胁迫应答过程中WRKY转录因子对ROS的调控网络,拓展采后香蕉果实冷胁迫响应的分子机制,并为采后香蕉果实贮运技术的改进提供新思路。

Postharvest banana fruit is highly susceptible to chilling injury during storage and transport time, which results in quality deterioration and commodity value loss. Reactive oxygen species (ROS) and WRKY transcription factors play important roles in plant stress response, however, whether WRKY transcription factors are involved in the regulation of ROS metabolism and their specific regulation mechanism when harvested bananas are exposed to chilling stress, remains largely unknown. In this project, on the base of physiological changes of ROS biosynthesis and elimination during chilling stress response in postharvest banana fruit, WRKY transcription factors and the downstream functional genes of ROS metabolism will be obtained according to banana fruit transcriptome database. The crucial WRKY transcription factors which regulate ROS metabolism-related genes will be selected and identified through DNA-protein interaction technologies. Moreover, the interacting proteins of key WRKY transcription factors will be characterized by protein-protein interaction approaches, and their influences to WRKY transcriptional regulation will be further analyzed. Thus, this project aims to systematically elucidate the regulatory networks of WRKY transcription factors on ROS metabolism during chilling stress response of postharvest banana fruit, advances our understanding of the molecular mechanism underlying chilling stress response in postharvest banana fruit and provides new cues for improving technologies during postharvest banana fruit preservation period.
