皮肤创面组织再生修复的有序调控与干预是当前研究的热点和难点。表皮干细胞(EpSCs)的数量和增殖能力不足是导致创面迁延不愈的最重要的原因之一。慢性创面高氧化应激和持续炎症的微环境,是导致EpSCs增殖能力受损的重要原因。因此,基于修复再生机制,设计新型生物材料,促进EpSCs增殖,形成经激活内源性再生能力与调控微环境,有望实现慢性创面原位再生修复。我们课题组研究发现,P311参与创面修复的整个过程,是调控EpSCs增殖的新分子。基于此,我们创新性的设计了搭载P311表达质粒的超小铜纳米酶,通过清除过量ROS降低氧化应激,减轻炎症,重塑有利于EpSCs增殖的微环境,同时通过基因治疗提供大量P311刺激EpSCs增殖;探索其可能的关键促进组织再生的通路Wnt/P38 MAPK-JNK-cJun。本项目的完成将为后续慢性创面的临床治疗,提供一种新的促进皮肤组织有序再生修复的干预靶点和手段。

The orderly regulation and intervention of skin wound tissue regeneration and repair is the hotspot and difficulty of current research. The insufficient number and proliferation of epidermal stem cells (EpSCs) is one of the most important reasons for the delayed wound healing. The high oxidative stress and persistent inflammation microenvironment of chronic wound are the most important reasons for the impaired proliferation of EPSCs. Therefore, based on the mechanism of repair and regeneration, new biomaterials are designed to promote the proliferation of EpSCs and form an activated endogenous regeneration ability and regulatory microenvironment, which is expected to realize the in situ regeneration and repair of chronic wounds. Our research group found that P311 participates in the whole process of wound repair and is a new molecule regulating the proliferation of EpSCs. Based on this, we innovatively designed an ultrasmall copper nanoenzyme carrying P311 expression plasmid to reduce oxidative stress, alleviate inflammation, reshape the microenvironment conducive to the proliferation of EPSCs, and provide a large number of P311 to stimulate the proliferation of EPSCs through gene therapy, and further explore its possible key pathway Wnt/p38 MAPK-JNK-cJun to promote tissue regeneration. The completion of this project will provide a new intervention target and means to promote the orderly regeneration and repair of skin tissue for the follow-up clinical treatment of chronic wounds.
由“伤”和“病”导致的皮肤组织缺损严重影响患者生活质量甚至生命,如何实现组织重建与创面修复所需的有序调控与干预是是烧伤、创伤、整形与修复重建外科学面临的最为重要的临床问题。创面高氧化应激和持续失控的炎症微环境,导致表皮干细胞(ESCs)增殖迁移能力受损,是创面迁延不愈的最重要的原因之一。基于修复再生机制,设计新型生物材料改善微环境,形成经激活内源性再生能力与调控微环境,促进ESCs增殖迁移,有望实现慢性创面原位再生修复。本项目创新性地基于磁共振成像技术研发了可视化实时监测创面炎症程度的纳米诊断制剂,进一步针对炎症介质ROS、MMP酶和免疫细胞如巨噬细胞,研发了系列高效改善创面炎症免疫微环境的功能生物材料,率先实现创面炎症程度的可视化、可跟踪、可反馈,高效精准调控。在上述基础上,提出了促进功能性修复和炎症调控并重的治疗策略,构建了系列活性生物材料改善局部炎症微环境,同时释放P311分子或者在创面原位高表达P311分子,协同促进ESCs增殖迁移,加快创面再上皮化,促进创面原位再生修复。本项目的完成将为慢性创面的临床治疗提供一种新的促进皮肤组织有序再生修复的干预靶点和手段。相关成果在Advanced Materials、Nano-Micro Letters、Nano Today、Acta Pharmaceutica Sinica B、Acta Biomaterialia等高水平期刊发表13篇;申请国家发明专利5项;项目承担人在此项目的资助下,成功获得了国家自然基金委优秀青年科学基金、重庆市杰出青年科学基金、重庆市学术技术带头人、王正国创伤医学创新奖,重庆市沙坪坝十佳科技青年奖;培养博士2名,硕士生5名;培养了2位硕士学位的科研助理成功晋升为副研究员,均成功获批国家自然科学基金青年基金。
