我国猪集约化生产水平较低,主要表现是猪早期胚胎丢失率高,产仔数低,增加了资源消耗,制约了产业发展。而猪卵母细胞成熟质量是胚胎发育的关键。褪黑素(MT)和瘦素(Leptin)都是影响雌性动物生殖的重要因子。本课题组前期研究结果表明,MT通过上调Leptin影响猪的卵母细胞成熟、激素合成和产仔数。基于以上发现,结合体内实验和体外卵母细胞培养模型,首先明确MT和Leptin在卵巢中的合成和互作位点;然后通过前期预测和转录组、代谢组学分析结果,验证MT调控Leptin的介导因子,并研究所获得因子在卵母细胞成熟过程中的功能;最后通过回补和添加实验,筛选和发掘能够改善猪繁殖力的有效靶点。不仅揭示MT调控Leptin促进卵母细胞成熟的机制,更对生产中提高猪繁殖力具有实践指导意义。
英文摘要
The intensive production level of pig is low in China. It is mainly due to the early embryo lost which leads to low litter size and productive efficiency. Meanwhile, the quality of pig oocyte maturation is the key to embryonic development. Melatonin and Leptin have been proved to play important roles in female reproduction. In previous studies, we discovered that Leptin mediates the effects of melatonin on female reproduction in pig. Melatonin-Leptin pathway regulates oocytes maturation, steroid secretions and litter size. This study aimed to investigate the mediators of the Melatonin-Leptin regulation. For this purpose, by combination of in vivo experiments and in vitro oocyte culture models, we first locate the synthetization and interaction sites of Melatonin and Leptin signaling. Secondly, we will screen the mediators involved in Melatonin-Leptin regulation by Pre-predictive and transcriptome, metabolomics analysis results. The effects of the selected regulation mediators on oocyte maturation will be examined. By supplementary treatment of the selected mediators, we will further search for the possible molecular targets which can be applied to improve the reproduction efficiency in stock farming. Our findings from this study will not only allow us to understand the mechanism of MT-Leptin signaling in promoting oocytes maturation, but also provide a useful model for improving farrowing rate.
