喉癌是头颈部常见的恶性肿瘤,对化疗不敏感,手术会影响患者生活质量。“肿瘤干细胞”理论是肿瘤学研究的新突破口。但目前喉癌干细胞研究仍处于起步阶段。本课题组利用干细胞耐药性,在动物模型体内经过多代次筛选,获得的喉癌干细胞(m3细胞),具有极强的干性和耐药性。将筛选后的喉癌干细胞测序,选择高表达的淋巴细胞抗原6家族成员D(LY6D)作为研究对象。在107例喉癌样本中,PCR检测出LY6D呈高表达,且和病理T分期、临床分期、颈淋巴结转移存在相关性。LY6D还可以增加喉癌细胞对化疗药的耐药性。研究表明,ABC转运蛋白家族可提高肿瘤细胞耐药性,LY6D与该家族中的ABCB11存在相互作用。但目前尚无LY6D与ABCB11互作关系的具体报道。我们将研究LY6D和ABCB11的作用关系,分析LY6D引发喉癌干细胞耐药的机制。本课题从喉癌干细胞角度入手,研究喉癌的耐药机制,为喉癌靶向药物研发提供支持。

Laryngeal cancer is a common malignant tumor of the head and neck. It is not sensitive to chemotherapy, and surgery will affect the quality of life of patients. The "tumor stem cell" theory is a new breakthrough in oncology research. But laryngeal cancer stem cell research is still in its infancy. The research team used stem cell chemo-resistance, and obtained laryngeal cancer stem cells (m3 cells) through multiple generations of screening in animal models, which have extremely strong stemness and drug resistance. The screened laryngeal cancer stem cells were sequenced, and the highly expressed lymphocyte antigen 6 family member D (LY6D) was selected as the research object. In 107 cases of laryngeal cancer samples, LY6D was highly expressed by PCR and correlated with pathological T stage, clinical stage, and cervical lymph node metastasis. LY6D can also increase the resistance of laryngeal cancer cells to chemotherapy drugs. Studies have shown that the ABC transporter family can increase tumor cell resistance, and LY6D interacts with ABCB11 in this family. However, there is no specific report on the interaction between LY6D and ABCB11. We will study the relationship between LY6D and ABCB11, and analyze the mechanism of LY6D triggering drug resistance in laryngeal cancer stem cells. This project starts from the perspective of laryngeal cancer stem cells, studies the drug resistance mechanism of laryngeal cancer, and provides support for the development of targeted drugs for laryngeal cancer.
