Th17/Treg cell imbalance plays a pivotal role in the pathogenesis of rheumatoid arthritis (RA). Our previous studies have demonstrated that selective agonists of canabinoid receptor 2 (CB2) effectively suppressed synovitis and joint destruction in mice with collagen II induced arthritis (CIA). Based on our preliminary findings, the samples of synovium, spleens and lymph nodes were isolated from CIA mice after chronic treatment with a CB2 agonist. Compared with vehicle treated mice, the changes in the amount of Th17 cells, Treg cells, specific transcription factors, cellular products and expression profile of microRNAs (miRNA) in the samples from CB2-agonist treated mice were analyzed with immunohistochemistry, quantitative PCR, Western blotting, flow cytometry or RNA sequencing. In vitro, differentiation of Th17 cells and Treg cells were induced under polarizing conditions, respectively. The potential effects of a CB2 agonist on differentiation, expansion and functions of Th17 cells and Treg cells are determined. Inhibition of STAT3 activation, over-activation of STAT3, down-regulation and up-regulation of candidate miRNAs, or knock-down of SOCS3 were performed by various plasmid transfection to study whether the SOCS3/STAT3 pathway or candidate miRNA mediates the modulatory effects of the CB2 agonist. Dual-luciferase reporter assay verified whether SOCS3 or STAT3 is the target of candidate miRNAs. By completing this study, it would be clarified whether CB2 agonists restore Th17/Treg cell imbalance in RA, and whether the miRNA/SOCS3/STAT3 pathway mediates the therapeutic effects.
类风湿关节炎(rheumatoid arthritis, RA)是以慢性滑膜炎症和骨侵蚀为特征,最终导致关节畸形。本项目旨在研究CB2受体激动剂对RA的治疗作用及机制,以探索潜在的RA治疗新靶点。.本课题首先采用佐剂性关节炎(AIA)作为RA的动物模型,证实了CB2激动剂HU-308对不仅可减轻AIA小鼠的关节肿胀、滑膜炎症,也可减轻关节结构破坏,进一步证实了CB2激动剂对RA具有治疗作用。从各实验组小鼠脾脏中分离免疫细胞,采用流式细胞术检测不同组小鼠脾脏中各种免疫细胞的数量或比例,结果显示AIA 小鼠的脾脏组织中, CD19+ B细胞和 CD8+T 细胞的比例减少,CD4+T 细胞的比例增加,且活化的 CD4+T 细胞和Th17 细胞的比例均显著增加,Th17/Treg细胞比值明显升高;HU-308治疗不影响CD4+T细胞数和活化的 CD4+T 细胞比例,但降低Th17 细胞水平并增高Treg 细胞比例;HU-308治疗后AIA小鼠脾脏中Th17/Treg失衡得到显著改善;HU-308治疗不影响Th1 和 Th2 细胞的比例以及 Th1/Th2 比值。.初始CD4+T细胞分别在Treg极化条件下或Th17极化条件下培养,TGFβ呈剂量依赖性地促进Treg细胞分化;HU-308在0.125~0.5 ug/ml TGFβ的培养条件下,均可以促进Treg细胞分化、增加其数量;HU-308可上调细胞内TGF-β的表达以及包括 CD25、IL-2、IL-10 和颗粒酶 B等与Treg细胞相关的其他功能性标志物的表达;HU-308不能增强Treg细胞对Teff细胞增殖的抑制性功能。HU-308显著抑制 体外Th17 细胞的分化,并抑制Th17 特异性转录因子 Ror-γt的表达,以及抑制Th17 细胞中致炎细胞因子 IL-17A 和 IL-17F的表达。Western blot 结果表明,HU-308 可诱导初始的 CD4+T 细胞内p-JNK/JNK 和 p-AKT/AKT 信号通路的轻度激活以及p-Smad2/Smad2、p-Stat5/Stat5显著上调。这些结果表明,HU-308 通过 JAK/STAT5 和 TGF-β/SMAD 信号通路调节 Th17/Treg 平衡。.通过这些研究,证明了CB2激动剂有望成为新型的RA治疗药物,并初步阐明了相关的作用机制。
