英文摘要
Carrots (Daucus carota L.), one of the most important root vegetables in the Apiaceae family, are cultivated worldwide. Carrots are mainly classified into two groups according to exact botanical determination: the carotene group (variety sativus) and the anthocyanin group (variety atrorubens). Carotene group members, also known as non-purple carrots, accumulate massive amounts of carotenoids in their roots; anthocyanin group members, also known as purple carrots, accumulate high levels of anthocyanins in their roots. Non-purple carrots are considered to have arisen from purple carrots that acquired mutations. Purple carrot root pigmentation extensively varies across different carrot genotypes, ranging from the purple peridermal carrot type (purple periderm but nonpurple phloem and xylem) to the solid purple carrot type (purple periderm, phloem, and xylem). . Two loci, P1 and P3 on chromosome 3, condition and modify the anthocyanin biosynthesis in taproots and leaves of carrot have been identified. We have characterized the functions two candidate genes, DcMYB113 and DcMYB7, which underlie the P1 and P3 loci, respectively. The DcMYB113 and DcMYB7 genes condition and modify the anthocyanin biosynthesis in taproot and leaf of carrot. The P3 locus hosts six different DcMYBs like genes, including DcMYB6 and DcMYB7. The precise regulation mechanism of the DcMYBs gene cluster at P3 locus on chromosome 3 in anthocyanin biosynthesis of carrot is not clear.. In this project, based on the previous results, we used the different genotypes of purple carrots, orange and wild-type carrots as materials. We mainly focus on the gene function of the DcMYBs gene cluster at P3 locus using methods of morphological, physiological, anatomical, gene clone, promoter clone, gene expression profiles, yeast hybrid system, transgenic plants with over-expression and gene stable editing using CRISPR/Cas9 system, OMICS analysis, etc. An extensive picture of the precise regulation mechanism of the DcMYBs gene cluster at P3 locus on chromosome 3 in anthocyanin biosynthesis of carrot shall be attempted. The project shall help to ascertain the function of DcMYBs gene cluster involved in precise regulation mechanism anthocyanin biosynthesis of carrot. The results will further enrich the molecular mechanism of carrot color formation, provide scientific basis for the evolution and domestication of carrot, and also provide basis for the improvement of carrot quality.
紫色胡萝卜是最早得到驯化的胡萝卜品种,在不同组织部位积累花青素。紫色胡萝卜肉质根花青素的合成受到3号染色体上P3位点基因簇的调控,该~500kb区域的基因簇由6个DcMYBs基因串联组成,精确调控胡萝卜肉质根、叶柄到花的花青素合成。本项目以不同花青素积累类型的胡萝卜为试验材料,筛选鉴定到分别调控胡萝卜紫色叶柄和紫色中心花性状形成的关键基因DcMYB11c和DcPCF,并探究了谷胱甘肽-S-转移酶介导的胡萝卜花青素合成后的转运机制。(1)共鉴定出146个胡萝卜R2R3-MYB转录因子并划分为19个亚家族,不均匀地分布在9条染色体。S6亚家族包含7个MYB家族成员。(2)在胡萝卜3号染色体上P3位点基因簇内鉴定获得MYB基因DcMYB11c,在紫色叶柄高度表达。在非紫色胡萝卜过表达DcMYB11c导致植株积累大量花青素。基因编辑 ‘深紫’DcMYB11c叶柄中花青素含量急剧下降。紫色叶柄胡萝卜材料中存在3个转座子,而绿色叶柄胡萝卜材料中不存在,这3个转座子起到转录激活作用,促进DcMYB11c在胡萝卜叶柄中高度表达,进而导致紫色叶柄的形成。(3)胡萝卜一朵或几朵紫色中心花出现在白色伞形花序中,DcPCF在紫色中心小花中高度表达,是决定中心花具有深紫色表型的关键基因。我们提出伞形花序中心花从紫色到非紫色的转变发生的3个事件,推测胡萝卜花是由一朵单独的花演化而来的。(4)DcGST1在紫色胡萝卜品种的紫色肉质根组织中高表达,而在紫色和非紫色胡萝卜品种的非紫色肉质根组织中几乎不表达。花青素激活因子DcMYB7与DcGST1启动子直接结合,激活其在肉质根中的表达。DcGST1和DcMYB7的共表达显著增加胡萝卜愈伤组织中花青素含量。综上所述,本项目进一步深入解析了胡萝卜3号染色体P3位点的MYB基因簇的DcMYB11c和DcPCF基因调控胡萝卜紫色叶柄和紫色中心花的花青素合成的机理,并揭示了DcGST1基因在胡萝卜花青素转运中的重要作用。项目结果为胡萝卜优异品种的创制及改良提供了借鉴,也为其它作物中花青素组织特异性积累的研究提供理论参考。
