T 细胞亚群间的平衡是维持免疫系统内部稳定的重要环节，抗肿瘤免疫尤以细胞免疫最为重要。（非小细胞）肺癌是常见的恶性肿瘤，其发生发展与机体T细胞免疫失衡密切相关，然而具体机制未明。TIM-1是一个重要的免疫共刺激因子，与其配体TIM-4相互作用，参与T细胞的分化、增殖，维持机体免疫稳定，但其在肺癌中的作用尚不明确。通过TGGA数据库分析表明TIM-4在肺癌组织中低表达。前期研究制备的TIM-1-Fc融合蛋白具有调控T细胞活化的功能。结合预实验和现有研究，我们假设“靶向共刺激分子TIM-1调控肺癌免疫失衡，促进机体抗肿瘤免疫应答。”为止，本项目拟以TIM-1为靶分子，以肺癌的免疫失衡为切入点，借助组织学、细胞生物学、免疫学、动物学手段，确证TIM-1-Fc靶向调控TIM-1及其信号途径，旨在阐释TIM-1基因对肺癌免疫失衡调控和抗肿瘤免疫的可能作用机制。此研究将为肺癌的防治提供新思路和新靶点。

The balance of T cell subsets is an important link to maintain homeostasis of the immune system, especially the cellular immunity is the most important part of anti-tumor immunity.Non-small cell lung cancer is a common malignant tumor. Its occurrence and development are closely related to the T cell immune imbalance. However, the specific mechanism remains unclear. TIM-1 is an important costimulatory molecule, which interacts with its ligand TIM-4 and participates in T cell differentiation, proliferation and immune stability. But its role in lung cancer is unclear. Low expression of TIM-4 was found in lung cancer tissues by TGGA database analysis. The TIM-1-Fc fusion protein prepared in previous studies has the function of regulating T cell activation. Combined with pre-experiments and existing studies, we hypothesized that "targeted costimulatory molecule TIM-1 regulates the immune imbalance of lung cancer and promotes the anti-tumor immune response of the body." To this end, we focus on the study of TIM-1 mediated lung cancer immune imbalance by histology, cell biology, immunology, animal model, which confirms TIM-1-Fc targeting regulation of TIM-1 and its signaling pathway, aiming to elucidate the possible mechanism of TIM-1 signaling pathway regulating immune imbalance of lung cancer and anti-tumor immunity. This study will provide new ideas and targets for the prevention and treatment of lung cancer.
