Late blight caused by Phytophthora infestans is a devastating disease in potato production. In-depth analysis of the immune response mechanism of potatoe is very important to the selection and breeding of potato disease resistant varieties. The effector protein secreted by P. infestans can target plant proteins to interfere with plant immune system to achieve infection. At present, the research on plant immune response mostly focuses on ubiquitination system and immune-related proteases etc. Whether there is a new immune response mechanism in plants remains to be resolved. Our previous study found that the potato ribosomal protein StRPS5 could interact with the P. infestans effector Pi16275. Using VIGS technology, it was found that the RPS5 silent tobaccos have greatly reduced resistance to P. infestans. In view of the fact that there are few studies on plant ribosomal proteins involved in immune response and these studies often lack molecular mechanism analysis, this project intends to use StRPS5 as an entry point to analyze the interaction mechanism between StRPS5 and Pi16275, and analyze the effect of knockout/silence and overexpression of StRPS5 gene on potato resistance to late blight, and then clarify the molecular mechanism of StRPS5 regulating potato immune response. The successful implementation of this project will reveal the molecular mechanism of ribosomal protein regulating plant immune response, further improve the research on plant immunity mechanism, and provide theoretical guidance for the selection and breeding of disease-resistant potato varieties.
