帕金森病（PD）是全球第二大神经退行性疾病，其发病机制目前尚未阐明。miRNA与PD密切相关，通过靶基因软件综合分析，发现miRNA-296-5p在蛋白质gasdermin D（GSDMD）呈高表达，疑似GSDMD为靶基因，且GSDMD是细胞焦亡过程中的关键物质。研究发现：补肾止颤方可降低PD大鼠中脑黑质区α-突触核蛋白（α-syn）的表达。我们提出假设：补肾止颤方基于miR-296-5p对GSDMD的调控，减少DA能神经元内α-syn的异常聚集，发挥治疗PD的作用。拟用RT-PCR、免疫组化、Western-blot、流式细胞术测定补肾止颤方干预前后miR-296-5p、IL-1β、GSDMD、Caspase-1、α-syn、TH表达变化及神经元细胞焦亡情况，采用荧光素酶报告基因技术测定miR-296-5p与GSDMD的靶标关系，以揭示补肾止颤方治疗PD的效应机制及作用环节。

Parkinson's disease (PD) is the second largest neurodegenerative disease in the world,and its pathogenesis has not yet been elucidated.MiRNA is closely related to PD,Through comprehensive analysis of target gene software,The miRNA-296-5p was found to be highly expressed in the protein gasdermin D (GSDMD), suspected to be the target gene of GSDMD,and GSDMD is a key substance in the process of cell pyrolysis. It was found that the expression of alpha-syn was decreased in the Bushen Zhichan Decoction in PD rats.We hypothesized that the regulation of GSDMD by miR-296-5p reduces the abnormal aggregation of alpha -syn in DA neurons and plays a role in the treatment of PD.RT-PCR, immunohistochemistry, Western-blot and flow cytometry were used to detect the expression of miR-296-5p, IL-1 beta, GSDMD,Caspase-1, alpha -syn and TH before and after Bushen Zhichan Fang intervention,and the focal death of neurons.Luciferase reporter gene technology was used to determine the target relationship between miR-296-5p and GSDMD, so as to provide an objective basis for the mechanism and role of Bushen Zhichan Fang in the treatment of PD.
