维吾尔族妇女宫颈癌晚期（盆腔淋巴结转移）患者多，预后差。肿瘤细胞脂肪酸代谢的重编程对细胞转移过程提供选择性优势。前期研究：肽基脯氨酰同分异构酶（Pin1）在宫颈癌组织和细胞系中高表达，可增加肿瘤细胞脂肪酸含量并促进细胞侵袭；下调Pin1后肿瘤细胞脂肪酸含量降低，可抑制细胞侵袭，Akt和PPARγ是其可能靶基因。推测：Pin1通过Akt /PPARγ调控脂肪酸代谢的重编程促进宫颈癌的转移。本项目拟以Pin1为关键分子，研究其与脂肪酸代谢相关信号通路（Akt/SREBP-1和PPARγ），脂肪酸代谢关键酶之间的相互作用，从组织、基因转染技术细胞和移植瘤动物模型等不同层面，采用双荧光素酶报告基因、免疫共沉淀等方法，系统分析Pin1调控宫颈癌细胞脂肪酸代谢和促进侵袭转移的作用机制，为进一步阐明宫颈癌发生与侵袭转移机制，以及宫颈癌治疗及预后判定等研究提供理论依据和实验基础。

Cervical cancer in Uighur women has a high morbidity and mortality. The advanced stage patients of cervical cancer in Uighur women are more than other patients.Reprogramming of Fatty Acid Metabolism in Tumor Cells Provides Selective Advantage for Cell Metastasis. Previous studies have found that Pin1 is highly expressed on cervical cancer cell, which increased the lipid content of tumor cells and also promote the invasion of tumor cells. Combined with the progress of peer research, it is speculated that: Pin1 may have an important impact on the invasion and metastasis of cervical cancer by reprogramming the fatty acid metabolites of cervical cancer cells. In this project, Pin1 is used as the key molecule to study its interaction with fattyacid metabolism related signaling pathways (Akt/ SREBP-1 and PPARγ) the interaction between key enzymes of fatty acid metabolism.The mechanisms of Pin1 regulating the fatty acid metabolism and promoting invasion and metastasis of cervical cancer cells were systematically analyzed using double luciferase reporter gene, immunoprecipitation method from different levels of tissue, gene transfection technology cells model and animal model of xenografts..To further elucidate the mechanism of the occurrence and invasion of cervical cancer and to provide an important theoretical basis and experimental basis for the prediction, treatment and prognosis of cervical cancer.
