不同来源的蜂胶化学组成差异较大,蜂胶产品质量良莠不齐、活性难以稳定发挥。近年来建立的多指标定量结合指纹图谱的方法在蜂胶真实性和质量一致性方面取得很大进展,但化学成分含量高低不能反映蜂胶在体内的生物学活性能力。本项目拟开展蜂胶体内动力学和代谢组学研究,运用LC-TOF-MS/MS等技术,建立大鼠血浆和尿液中蜂胶代谢组成分检测方法,考察大鼠灌胃给予蜂胶后的代谢动力学,探索蜂胶复杂体系可能的活性成分在体内代谢过程和代谢途径;筛选出在体内发挥作用的活性成分;建立基于化学成分分析和生物效应检测的综合量化集成质控新方法。本项目的开展将为蜂胶在体内发挥作用的物质基础研究奠定理论基础,建立的以“质量-活性-代谢”量化关系的综合指标评价蜂胶质量的方法与体内活性切实关联,同时能反映不同活性成分对蜂胶整体活性贡献度,补充和完善了现行“唯成分论”的蜂胶质量控制评价方法与标准,为蜂胶质量评价提供科学依据。

The chemical composition of propolis various greatly from different areas, the qualities of propolis product must be payed attention to their divergences. In recent years, due to the establishment of multi-index and fingerprint control methods, great progress has been made in the authenticity and quality consistency of propolis. However, the research on the correlation between propolis quality and in vivo activity has not been involved. This project intends to carry out the study of metabolic kinetics and metabolomics. The method for the determination of metabolic components of propolis in rat plasma and urine is developed by LC-Q-TOF-MS/MS and other techniques. The metabolic kinetics of various chemical components from propolis after intragastric administration in rats is investigated, and the possible metabolic processes and pathways of active components in complex propolis system are explored. Bioactive ingredients in propolis that play a role in vivo have been screened, and a new integrated quantitative quality control method based on chemical composition analysis and biological effect detection has been established. This project will lay a theoretical foundation for the material basis research of propolis in vivo, and the new method for propolis quality control by comprehensive index of quantitative relationship of "quality-activity-metabolism" is related to the activity in vivo, and can also solve the problem of the contribution of different active ingredients to the overall function of propolis, and supplement and improve the current "the theory of the unique importance of class origin" propolis. The evaluation methods and standards of quality control provide scientific basis for evaluating the quality of propolis.
