猪骨骼肌细胞增殖直接决定肌肉生长速度和产肉量,是影响产肉性状形成的重要因素。调节单元(regulon)是真核生物中由转录因子及其调控基因组成的一组基因集合,是精确剖析基因调控网络,解析复杂性状形成分子机制的关键。但目前相关研究在猪中尚未见报道。申请人及前人研究发现组织特异的调节单元在哺乳动物间是高度保守的。因此,本项目拟通过跨物种比较分析鉴定人、小鼠和猪间保守的骨骼肌细胞特异调节单元,构建骨骼肌细胞发育状态(静息、增殖、迁移和融合分化等)转换轨迹和调节单元动态表达图谱。结合猪骨骼肌卫星细胞增殖分裂不同时期转录组数据鉴定猪骨骼肌细胞增殖相关特异调节单元。利用CRISPR-dCas9技术在分子、细胞水平对调节单元基因功能进行验证。在常规育种群体中评估调节单元基因对猪产肉性状的遗传效应,为猪遗传育种改良提供新的候选基因。本项目能为解析动物复杂性状形成的分子机制提供新的研究思路。

The proliferation of pig skeletal muscle cells directly determines the speed of muscle growth and meat production and is an important factor affecting the formation of meat production traits. In eukaryotes, a regulon is a collection of a transcription factor and all its transcriptional target genes. It is the key to accurately dissect gene regulatory networks and analyzing the molecular mechanism of complex trait formation. However, related research has not yet been reported on pigs. The applicant and predecessors found that tissue-specific regulons are highly conserved among mammals. Therefore, this project aims to identify the conserved skeletal muscle cell-specific regulons among humans, mice, and pigs through cross-species comparative analysis, and construct skeletal muscle cell development trajectories (quiescent, proliferation, migration, fusion, differentiation, etc.) and dynamics expression atlas of regulons. Combining the transcriptome data of porcine skeletal muscle satellite cells at different cell cycles to identify specific regulons related to the proliferation of porcine skeletal muscle cells. By detecting the function of regulons at the molecular and cellular levels using CRISPR-dCas9 and evaluating the genetic effects of regulons on pig meat production traits in conventional breeding populations, this project will provide new candidate genes for pig genetic breeding improvement. This project can also provide new research ideas for the analysis of the molecular mechanism of the formation of complex animal traits.
