家蚕(Bombyx mori)是我国重要的经济昆虫,人工饲料饲养家蚕具有省力化、有利于防病等诸多优势,是蚕丝产业转型升级的重要途径。然而与传统桑叶喂养相比,人工饲料饲育的家蚕产丝量少、丝蛋白合成效率低的问题一直未得到解决。家蚕对营养物质的吸收利用是影响茧丝产量与质量的重要因素。已有研究证明肠道微生物能够为宿主提供生长所需的营养物质,然而家蚕肠道微生物是否促进家蚕的营养代谢以及对产丝量的影响尚不明确。本项目在对比不同饲养方式的家蚕肠道微生物肠球菌种群变化的基础上,培养、筛选、分离鉴定可能影响茧丝产量的关键差异菌株,回接验证特定菌种、菌株对产丝量等的影响,以代谢组学与细胞分子生物学技术对其可能的蚕丝蛋白合成、作用靶点与调节机制进行初步探究,揭示不同家蚕产业饲养模式下肠道微生物代谢-宿主-丝蛋白合成轴调控的初步机理,为蚕桑产业开发新型微生态制剂与应用提供理论支持。
英文摘要
Bombyx mori is an important economic insect in China. Raising silkworm with artificial diet has many advantages, such as labor-saving and disease-prevention, which is an important way to transform and upgrade the silk industry. However, compared with traditional mulberry leaf feeding, the problems of less silk production and low efficiency of silk protein synthesis in artificial diet have not been solved. The absorption and utilization of nutrients by silkworm is an important factor affecting the quantity and quality of cocoon silk. It has been proved that gut microorganisms can provide nutrients for the host, however, whether intestinal microorganisms promote the nutrient metabolism of silkworm and their effects on cocoon quantity is still unclear. In this project, the key differential strains of Enterococcus which may affect the silk production were screened, isolated and identified from the gut of silkworm on the basis of comparing the difference of intestinal microbiota in silkworm with different rearing methods. Then the isolated Enterococcus strains were re-introduced to silkworm and the effects of them on silk production were verified. Furthermore, the possible mechanisms about silk protein synthesis and its regulation were explored by metabolomics and molecular biology techniques. This project will provide preliminary mechanism of microbe metabolic -host -silk protein synthesis-axis and provide theoretical support for the development and application of novel microecological agents in the silkworm industry.
古往今来,蚕丝产业在我国经济发展和文化传播中扮演着重要角色。近年来随着产业升级和科学技术的发展,蚕丝拥有了新的产业方向,其在生物医药、电子科技及食品等领域的应用也越加广泛。人工饲料育蚕具有高效、可标准化、不受季节限制等优势,是蚕桑产业转型升级的迫切需要。然而,与桑叶饲养相比,存在生长慢、产丝量少等问题,如何提高人工饲料养蚕的产丝量是亟需解决的关键问题。肠道微生物在参与宿主的营养代谢、生长发育、免疫调节等方面发挥着重要作用。我们的前期研究发现人工饲料育蚕与桑叶育蚕相比,肠道微生物存在显著差异。本项目进一步从家蚕肠道中分离培养关键差异微生物,筛选出能够显著促进家蚕生长及产丝量的功能菌株并揭示作用机制,取得了系统性的重要研究成果及进展。主要包括:(1)从家蚕肠道中共分离培养关键微生物肠球菌(Enterococcus)15株,包括4个种,即蒙氏肠球菌E. mundtii,鹑鸡肠球菌E. gallinarum,铅黄肠球菌E. casseliflavus和海氏肠球菌E. hirae;(2)从以上菌株中筛选出1株能够显著促进家蚕生长及产丝量的菌株—鹑鸡肠球菌E. gallinarum ZZ-8,并探究了菌株的不同添加浓度对产丝量的影响,同时还发现菌株具有提高家蚕免疫力的潜力;(3)除肠球菌以外,还筛选到了另外1株能够显著促进家蚕生长、产丝量及抗病性能的益生菌戊糖片球菌Pediococcus pentosaceus ZZ61;(4)进一步通过肠道微生物多样性、非靶向代谢组学、基因组学等多组学联合分析揭示了益生菌影响家蚕生长及产丝量的作用机制,表明益生菌可能通过影响家蚕的营养代谢、调节肠道微生物和代谢产物进而促进家蚕生长和产丝量。本项目所获得的成果不仅加深了我们对益生菌如何影响宿主生理的认识,也为今后利用家蚕模型研究宿主-微生物互作提供了研究方向,同时也为益生菌在养蚕业中的应用提供了理论支持。
