从深海热液口到人体内都有分布的Epsilon-变形菌主要以微好氧和厌氧方式生存,因此氧气感知能力对它们适应环境变化发挥着重要功能。我们的生信分析发现这类细菌的基因组含有多拷贝的氧气感知蛋白—蚯蚓血红素蛋白。目前对该结构域感知氧气后引发的细胞应答过程包括它与其它结构域或蛋白的互作机制、信号转导模式和下游的信号输出的研究十分匮乏。我们在模式菌株中的初步研究发现一个含蚯蚓血红素结构域的蛋白与下游的转录调控因子存在共转录和蛋白-蛋白互作,是一种新型的氧气感知与应答调控模式。本项目拟对这两个蛋白开展功能研究,阐明两个蛋白之间的相互作用机制以及不同氧气浓度下两个蛋白如何调控靶基因表达作出生理响应。该研究将揭示一种新型的氧气感知与应答作用模式,为合成生物学的基因回路设计提供氧感知元件相关的理性设计原理,并且对Epsilon-变形菌纲的病原菌防治以及极端环境富集的细菌资源开发利用有重要指导作用。

Oxygen sensing plays an important role in the environmental adaptation of Epsilon-proteobacteria, which are mostly microaerophilic or anaerobic, widely distributed from deep-sea hydrothermal vent to human gut. Our comparative genomic analysis discovered that this group of bacteria encode multiple copies of oxygen-sensing domain such as hemerythrin. Several in vitro biochemical and structural analysis proved that this protein domain can bind oxygen. However, the cellular responses triggered by oxygen-binding of this protein remain elusive, including the interaction of this domain with other domains or proteins, signal transduction mode or signal outputs. To explore these questions, we studied a hemerythrin-containing protein and its cognate downstream regulator in a model strain. Our preliminary results indicate that these two proteins are co-transcribed and interact with each other, which are novel oxygen sensor and regulator pair. Thus, we propose to carry out functional studies on these sensor-regulator pair to elucidate their interaction mechanisms and how they affect cellular response to oxygen level. This study will reveal a new oxygen sensing and response mechanism, which will provide a useful oxygen sensor and working principles for gene circuit design in synthetic biology. Furthermore, this work will shed important insight into the bioprospecting of this group of bacteria abundant in extreme environment and also the control of bacterial pathogens.
本项目聚焦于Epsilon-变形菌纲的一种新型氧气感知与应答机制,全面探索了氧气感知蛋白(hemerythrin)与下游转录因子互作的作用模式。.Epsilon-变形菌纲是变形菌门中生态分布最广的小类群之一,覆盖从深海热液口到人类胃肠道等多样化环境。虽然其适应氧气变化的能力对生态适应性至关重要,但相关机制研究尚属空白。其中细菌的氧气感知蛋白hemerythrin因其结合氧气和铁离子的能力在 Epsilon-变形菌中广泛分布,且具有独特的CxC...CxxC motif,是研究其感知与应答机制的理想对象。通过对数据库中70余种全基因组测序菌株的比较分析,发现 Epsilon-变形菌中约83%含有hemerythrin,并呈现多拷贝分布(1~7个拷贝)。进一步的系统发育分析揭示了hemerythrin从共同祖先进化而来,其在细菌铁稳态调控及氧气感知中具有重要功能。在模式菌株 Campylobacter jejuni 81-176中,含CxC...CxxC motif的hemerythrin蛋白herB与 herR(转录因子)基因相邻排布且为共转录蛋白。双杂交、免疫共沉淀及突变实验明确了HerB和HerR的互作关系,且证明HerB的C端是关键互作区域。转录组分析显示,HerB-HerR协同调控了多条铁转运系统的转录水平,对细菌铁稳态和适应氧气变化起重要作用。此外,HerB-HerR还显著下调了生物膜形成相关基因cprRS 的表达,从而抑制生物膜的形成。本研究首次揭示hemerythrin与转录因子协作调控氧气应答和铁稳态的新模式,填补了细菌氧气感知领域的研究空白。阐明Campylobacter jejuni等病原菌的氧气适应机制,为抗菌药物开发提供靶标;对深海极端环境中的 Epsilon-变形菌资源利用提供了理论支持。.本基金项目支持发表SCI论文2篇(一区期刊1篇,2区文章1篇),其中近期接收在PNAS上的文章系统描述了作为氧气感知蛋白hemerythrin模式菌株的空肠弯曲杆菌中鞭毛系统上的FlgX蛋白结构和生化特征,细菌鞭毛的定子旋转是一类旋转分子马达,现在已被认为在生物系统中有更广泛的应用,而该蛋白的研究为促进定子旋转的辅助蛋白提供了新的见解。同时,我们还开发了稳定表达外源蛋白的筛选方法,在此期间项目负责人参与培养硕士研究生1名。
