研究认为多囊卵巢综合征(PCOS)源于胚胎时期宫内高雄激素的刺激,继而引起生殖系统在表观遗传学水平发生重新编程,最终导致出生后PCOS的发生,表观遗传机制参与PCOS的发病机制并对子代健康产生影响。深入研究PCOS的表观遗传效应,寻找有效的预防措施,具有重大意义。我们前期研究发现养精种玉汤可改善PCOS大鼠子代高雄激素血症,本课题拟以大鼠孕期高雄激素环境构建类人的PCOS妊娠模型,应用经典古方养精种玉汤合寿胎丸干预,采用焦磷酸测序、行为生物学等技术观察,旨在探讨养精种玉汤合寿胎丸能否有效改善母系胎盘表观遗传效应和胎盘的功能,预防或者减轻雌性子代生殖代谢和情志异常的症状,降低子代疾病发生的风险。该研究结果可望为中药预防PCOS发病提供实验依据,并将有助于丰富中医“治未病”的科学内涵。
英文摘要
Many studies have suggested that Polycystic ovary syndrome (PCOS) is a fetal-derived disease, which stems from the stimulation of intrauterine hyperandrogen in the embryonic period, which in turn causes reprogramming of the reproductive system at the epigenetic effect during fetal development until offspring. The pathogenesis of PCOS and the health of offspring may be related to epigenetic modifications. Therefore, it will be of great significance to in-depth study of the epigenetic effects of PCOS and the search for preventive treatments. Our previous study have suggested that chinese medicine formula Yangjingzhonguyu could reverse the hyperandrogenism of PCOS-like rats. This study aim to explore how maternal androgen excess in late pregnancy ameliorate placenta function and its epigenetic, and regulate of fetal programming, especially in female offspring, moreover, to evaluate the effects of chinese compound formula Yangjingzhongyu and Shoutai Pillduring pregnancy in PCOS-like rats model. Of note that PCOS with hyperandrogenism induced epigenetic transgenerational inheritance of reproductive, metabolic and mental disorder via the network of environmental factors - placental epigenetic effect - progeny reproductive metabolism. Yangjingzhongyu and Shoutai Pill may improve the epigenetic effect of the maternal placenta, prevention or alleviate the symptoms of reproductive metabolism and emotional changes in female offspring. Hence, to improve the environmental factors of PCOS during pregnancy, it is a best way to prevent PCOS, and reflect the advantages of TCM "prevention of disease", "prevention is better than treatment" and "eugenics".
