木材是重要的可再生自然资源，为多种工业提供原料。杨树是我国主要的用材树种，在保障木材安全和生态安全中发挥关键作用。杨树细胞壁中阿拉伯半乳聚糖蛋白（AGP）对于次生壁发育和木材形成过程中起重要作用。我们通过生物信息学方法寻找杨树木质部中参与AGP合成的糖基转移酶，运用生化方法鉴定其中三个GT14，GT31和GTNC蛋白分别参与AGP中葡萄糖醛酸、半乳聚糖和鼠李糖的合成；同时使用CRISPR-Cas9技术对这三个基因进行敲除，分析AGP不同糖基对于木材材性的影响。本研究结果有助于阐明木材形成中复杂的生物学过程，为树木分子育种提供理论依据和技术指导。

Wood is an important renewable natural resource that supplies raw materials for a variety of industries. Poplar is the main timber species in China and plays a pivotal role in ensuring the strategic timber safety and ecological balance. The cell wall glycoprotein AGPs play important regulatory roles in secondary wall development and wood formation. The project aims to identify xylem highly expressed glycosyltransferases involved in AGP biosynthesis using bioinformatic tools. Three glycosyltransferases from family GT14, GT31 and GTNC are characterized with GlcAT, GalT and RhaT activities respectively. The three GTs are knocked out using CRISPR-Cas9 technique and their biological effects on wood formation and quality are analyzed. The results of this study are helpful in understanding the complex biological processes in wood formation and providing theoretical basis and technical support for tree molecular breeding.
