脑缺血再灌注损伤是决定脑中风患者生存和预后的重要环节,如何促进再灌注损伤后血管新生、改善血供是改善患者预后的临床难题。眼针带针康复临床效果显著,文献表明再灌注后缺血半暗带中存在侧支循环,有神经元存活。前期研究眼针带针能够上调MCAO模型大鼠脑缺血半暗带区域内VEGF、VEGFR1、VEGFR2mRNA及蛋白表达水平,增加Ang1与Tie2结合,因此推测眼针带针对MCAO大鼠可能有促进血管新生作用。VEGF/VEGFR是新生血管必须调节信号通路,VEGFA起着重要作用,调节细胞的迁移、稳定血管结构。那么眼针带针康复是否通过调控VEGFA/VEGFR2信号通路发挥其促进血管新生、建立侧支循环作用呢?故提出假说,课题将观察眼针带针康复对MCAO模型大鼠血管新生内皮细胞迁移、稳定性蛋白及VEGFA/VEGFR2通路影响,探讨其促进血管新生、建立侧支循环机制,为其广泛推广奠定实验基础。
英文摘要
Cerebral ischemia-reperfusion injury is an important link that determines the survival and prognosis of patients with cerebral stroke. How to promote angiogenesis and improve blood supply after reperfusion injury is a clinical problem to improve patients' prognosis.The eye-acupuncture therapy combined with rehabilitation training is effective in the treatment of stroke. As reported, the cerebral ischemia reperfusion injury is an important pathophysiological process of stroke.Papers showed that ischemic penumbra occured after cerebral ischemia-reperfusion, and a large number of neurons survived in the collateral circulation. Our group proves that VEGF、VEGFR1、VEGFR2 expression and the level of Ang1 of MCAO model mice treated by the eye-acupuncture therapy combined with rehabilitation training can be increased and promoted the combination of Ang1 and Tie2. So that the eye-acupuncture therapy combined with rehabilitation training for MCAO model mice may have the function of coronary collateral circulation.Therefore,it is suggested that "the mechanism of effect of treatment of cerebral ischemia-reperfusion injury with eye acupuncture combined with rehabilitation training may be realized by VEGFA/VEGFR2 signaling pathway.”This study observe the proliferation, migration and stability proteins of neovascular endothelial cell, as well as the expression of VEGFA/VEGFR2 signaling pathway in MCAO modeled mice. If this project accomplishes,it would provide the theortical foundation for the spread of eye-acupuncture therapy combined with rehabilitation training .
