引导性骨再生(GBR)技术是当前针对牙槽骨缺损的常见治疗方法。其中引导性骨再生膜对于GBR手术效果有重大影响。目前临床中常用的GBR膜往往缺乏良好的生物屏障功能,且降解速率过快,不利于组织的再生修复。近年来Janus的不对称双面概念被应用于生物医学材料,本研究计划利用静电纺丝技术,设计并制备同时具有软组织屏障、抗菌能力和成骨/成血管活性的多功能Janus膜材料:以丝蛋白(SF)/壳聚糖(CS)电纺纳米纤维为内表面,并掺入负载Wnt/β-连环素激动剂-香草酸甲酯(MV)的明胶还原氧化石墨烯(GOG),抗菌的同时可促进成骨;以定向电纺的SF/CS纳米纤维作为外表面,形成屏障作用。本研究拟探索Janus纳米纤维膜的构建与表征,通过体外生物学实验研究其屏障能力、成骨/成血管以及抗菌性能;体内以大鼠动物模型评价Janus膜的生物相容性以及快速修复骨缺损的能力,为引导性骨再生膜的材料改进提供新的方向。

Guided bone regeneration (GBR) technology is currently a common treatment for alveolar bone defects. Guided bone regeneration membrane has a significant impact on the effect of GBR surgery. At present, GBR membranes commonly used in clinical situations are not conducive to tissue regeneration and repair because they often lack good biological barrier function, and express fast degradation rate. In recent years, Janus asymmetric double-sided concept has been applied to biomedical materials. This research plans to use electrospinning technology to design and prepare a multifunctional Janus membrane material with soft tissue barrier, antibacterial ability and osteogenic/angiogenic activity: Silk fibroin(SF)/chitosan(CS) electrospun nanofibers are prepared to form the inner surface and mixed with gelatin-reduced graphene oxide (GOG) loaded with Wnt/β-catenin agonist-methyl vanillate(MV),which possesses both antibacterial and osteogenic function; oriented electrospun SF/CS nanofibers are prepared as the outer surface to form a barrier. This study intends to explore the construction and characterization of Janus nanofiber membranes, In-vitro biological experiments are conducted to study the barrier function, osteogenic/angiogenic and antibacterial properties; in vivo rat animal models are used to evaluate the biocompatibility and ability to quickly repair bone defects of Janus membranes. This research is aimed at providing a new direction for the improvement of guided bone regeneration membrane materials.
