粘体动物是寄生于鱼类的后生动物寄生虫,其起源与进化一直存在诸多争议。目前,粘体动物属于刺胞动物的观点已被广泛认可,但其在刺胞动物内的系统发育位置尚不清楚。刺丝囊是刺胞动物的重要分类依据之一,不同刺胞动物的刺丝囊存在显著差异。粘体动物的极囊与刺丝囊在发育过程、结构及功能等方面极其相似,且有共同功能蛋白基因,研究二者的亲缘关系对阐明粘体动物在刺胞动物内的系统发育位置具有重要意义。本项目以极囊为切入点,通过“手术解剖”法获得纯化极囊,借助多组学分析平台,获得极囊蛋白质组数据,结合已知极囊和刺丝囊的蛋白质组,开展极囊和刺丝囊的比较蛋白组学、关键蛋白、同源基因簇等方面的研究和数据分析,探讨极囊与刺丝囊的亲缘关系及在刺丝囊中的进化地位,解析细胞器进化与生物进化的关系,阐明粘体动物与刺胞动物的亲缘关系及其在刺胞动物内的具体位置,为研究粘体动物的起源与进化提供新思路。
英文摘要
Myxozoans are metazoan parasites of fish and its origin and evolution have been controversial. Currently, it is well acknowledged that myxozoans are cnidarian. However, their phylogenetic position within the cnidaria still remains unclear. The nematocyst is one of the most important classification criteria of cnidaria. The nematocysts of different cnidarians have significant differences. The polar capsules of myxozoans are highly similar to the nematocysts of cnidarians in the development process, structure and function. They also have common functional protein genes. Studying their relationship is important for elucidating the phylogenetic position of myxozoans in the cnidaria. Taking the polar capsule as the entry point, the purified polar capsules were obtained through the "surgical anatomy" method. the proteomic data of polar capsules were also obtained with the help of the multi-omics analysis platform. Then we combined the know proteomic data of polar capsules and nematocysts, conducted analyses including comparative proteomics, key proteins, and homologous gene clusters, to explore the relationship between polar capsules and nematocysts and the evolutionary position of polar capsules within nematocysts, to explore the relationship between organelle evolution and biological evolution. We aim to clarify the relationship between myxozoans and cnidarians, and the specific position of myxozoans in cnidarians. The study will provide new ideas for solving the problem of myxozoan origin.
本项目以粘体动物/刺胞动物的极囊/刺丝囊为切入点,采用多组学方法,开展了极囊和刺丝囊的比较蛋白组学、关键蛋白、同源基因簇等方面的研究,主要结果如下:(1)获得了7种粘体动物的基因组学和转录组学数据,建立了粘体动物的组学数据库;(2)基于基因组和转录组学研究了粘体动物极囊毒液基因的多样性和起源与进化,发现粘体动物极囊毒液基因的多样性大大减少,其进化模式为趋异进化,与自由生刺胞动物的毒液在分子结构、预测功能和选择压力上存在显著差异;(3)基于单拷贝核基因开展了粘体动物的系统发育地位与发生年代分析,发现(粘体动物+螅型多足虫)+水母亚门的拓扑结构得到了确定且稳定的支持,建议粘体动物与螅型多足虫共同组成一个亚门。(4)分离了3种代表物种的极囊,发现热、尿素和氨处理能够触发大多数孢子和离体极囊的极丝释放,而NaCl、乙酸、乙醇、碳酸氢钠和CaCl2则不能触发极丝释放,重金属处理后可显著降低极囊的释放率;(5)开发了“定制综合蛋白质组参考数据库(customized comprehensive proteomic reference database)”,提供了全面、无污染的蛋白组学参考数据库;(6)比较蛋白组学证明了粘体动物极囊与刺胞动物刺丝囊的同源性,并通过透射电镜证据以及粘刺丝囊特有蛋白推测极囊应属于刺胞动物中的粘刺丝囊;(7)发现了刺丝囊的异质性组成及去中心化进化,揭示物种的特异表型创新模式,证明刺丝囊是粘体动物、刺胞动物成功适应性进化的基石;(8)进化基因组学研究发现了粘体动物的马赛克进化(mosaic evolution)模式,阐明了粘体动物与刺胞动物的亲缘关系及其在刺胞动物内的具体位置;(9)开展了粘体动物的物种多样性调查,发现了新种6个,丰富了各物种形态学数字影像和分子序列等信息。发表项目标注文章11篇,其中第一标注论文10篇,授权发明专利2项,培养博士2名,硕士2名。
