英文摘要
The first cleavage is the beginning of embryonic development and requires the cooperation of sperm and egg, but the contribution of sperm is rarely studied. As testis and sperm specifically expression protein, SSP411 was first reported by our team in the world on 2004. Lack the expression of SSP411, causes sperm abnormalities and fertilized eggs first cleavage delay. That means this protein is important in sperm and embryonic development. We construct the HA-tag transgenic mouse with CRISPR/CAS9 to trace protein detection, and construct the gene knockout mouse model with the same technology defects of the original knockout mouse. Based on the above mouse models, we propose that SSP411 is a component of epididymis sperm-specific cytoskeletal protein in MTOC, which plays an important role in the first cleavage of fertilized eggs. We are planning to express SSP411 protein and co-localization with MTOC in gametes and embryos of each stage using the HA-tag transgenic mouse model, to prove that SSP411 is a component of MTOC of fertilized eggs derived from sperm. Then, we are going to use SSP411 gene knockout model to prove one of the mechanism of the first cleavage delay is lack of SSP411 protein. Through the above research, we are going to analyze the SSP411 protein function for proving: sperm is necessary for MTOC to perform the cleavage function in fertilized egg, and sperm proteins contribute to the first cleavage in fertilized egg.
Ssp411基因(Sperm-specific protein 411)是本团队首次报道的,在哺乳动物睾丸组织中特异表达的基因,又被称为Spata20或SPATA20基因。已有研究显示,Ssp411蛋白主要存在于减数分裂后的圆形精子和长形精子中。本项目通过CRISPR/Cas9基因编辑技术,成功构建了Ssp411HA/HA标签小鼠模型和Ssp411-/-突变小鼠模型。借助Ssp411HA/HA小鼠模型,成功在成熟精子的顶体后鞘区域鉴定到Ssp411蛋白的表达;结合两种小鼠模型,我们发现Ssp411蛋白缺失表达,导致小鼠部分精子的精子环结构缺失,并形成不动或运动能力差的精子,这可能是雄性Ssp411-/-小鼠不育的原因。此外,Ssp411蛋白缺失表达可导致Septin-4蛋白在精子头部异常“堆积”,导致卵裂异常和囊胚率显著降低,影响早期胚胎发育。因此,SSP411基因可能是男性不育筛查和治疗的潜在靶点,对于因Ssp411蛋白缺失表达而发生精子鞭毛畸形的患者,可考虑抑制Septin-4蛋白活性的ICSI方案,以提高优质胚胎率和囊胚形成率,最终获得较好的妊娠结局。
