准确重建生物类群的系统发育是揭示生命体起源与演变动态、阐明生物多样性形成机理的基础。胡桃科是重要的阔叶林组成树种,间断分布于北美、欧洲和亚洲,少数种类延伸至美洲热带地区。其演化与分布格局复杂,孑遗成分多,化石资料丰富,是揭示北半球植物类群间断分布格局成因、探究北半球植物区系时空演变动态的关键类群。针对其系统发育关系不清、生物地理历史不明的问题,本课题基于良好的前期积累,拟采用强大的探针杂交捕获测序技术构建胡桃科高分辨率系统发育树,以之为基础推算各类群分化时间,分析物种多样化速率,重建祖先分布区,整合形态学、系统学、地理学等多领域研究成果,揭示胡桃科的时空演化历史。通过本课题的实施,可进一步验证北方热带植物区系等有关间断分布成因的假说,拓展对北半球区系演变的认知,研究成果也为全球气候变化背景下森林树种的保育实践提供基础和依据。
英文摘要
Accurately reconstructing lineage phylogeny is crucial for revealing the origin and evolution of organisms and elucidating the underlying mechanism of biodiversification. Juglandaceae, an important component of broad-leaved forest, areh disjunctly distributed in North America, Europe, and Asia, with a few species extending to tropical America. The complex pattern in evolution and distribution, abundant relic components of the Tertiary, and diverse fossil records make Juglandaceae a key group to reveal the underlying mechanisms of the intercontinental disjunct distribution pattern and the dynamics of the temporal and spatial evolutionary history of the Northern Hemisphere flora. Aiming at clarifying the unclear phylogeny and obscure biogeographic history, we will use the powerful Hyb-Seq method to reconstruct the phylogeny of Juglandaceae, estimate clade divergence time, analyze species diversification rate, and to reconstruct ancestral distribution range, and finally elucidate the biogeographic history of Juglandaceae. Our study will further test the hypotheses about the mechanisms of disjunct distribution such as the Boreotropical, broaden our understanding of the evolution of the Northern Hemisphere flora, and contribute to the conservation of woody plants under the background of global climate change.
重建生物类群的系统发育关系,追溯其起源与演化历程是生命科学的热点内容。北半球木本特有种和孑遗种类众多,具有重要的生态、经济和学术价值。古老的起源时间、复杂的演化格局、丰富的孑遗成分、多样的化石资料,让胡桃科成为北半球间断分布木本植物的典型代表,是深入开展系统发育研究、解析北半球植物区系时空演变动态、验证北方热带植物区系等假说的范式类群。本研究以重测序数据为基础,特别是在获得了最为关键的中南美洲类群材料的基础上,重建了基于核基因、质体基因的进化树,设置适宜化石标定策略,开展胡桃科系统发育基因组学与生物地理学研究。结果表明,胡桃科可分为马尾树亚科(含1属1种)、黄杞亚科(含4属15种)和胡桃亚科(含5属47种),黄杞亚科美洲分支的坚黄杞属与雀鹰木属关系不明,胡桃亚科的3个属(胡桃属、枫杨属和青钱柳属)存在核质冲突现象,胡桃属与山核桃属的物种树与基因树冲突严重;分化时间与生物地理分析显示胡桃科在白垩系起源于北美洲中、低纬度地区,在K-Pg boundary、始新统、中新统等古气候极端变化和特迪斯海、图尔盖海峡的消失等古地质事件的综合影响下,胡桃科在亚科、属和物种等不同尺度发生了谱系形成、灭绝和生态位分化。东亚受第四系冰川影响最小,加之喜马拉雅山脉的隆起和亚洲季风的增强进一步塑造了良好的环境,亚洲胡桃科类群得以最大程度的保存并繁衍,进而形成了北美化石类群古老且种类多、东亚现存物种丰富度高的分布格局。在项目实施过程中,还在广西发现了海南特有胡桃科物种——海南黄杞,为珍稀树种的保护提供了基础数据。通过本项目的实施,培养了硕士和博士研究生,发表了研究论文,实现了预期目标,取得了良好的成果。
