核酸和蛋白质的高灵敏检测对于生命科学研究和疾病的早期诊断具有重要意义。传统检测技术因灵敏度低、操作程序复杂、成本昂贵,无法满足科研或临床实验室的需求。CRISPR/Cas12a检测系统具有简单、快速、灵敏、廉价等优势,但由于受到PAM识别位点的限制,难以在核酸和蛋白检测中广泛应用。本项目在前期工作基础上,拟通过探索PAM位点的识别机制,对CRISPR/Cas12a系统进行改造和优化,克服PAM位点的依赖;结合邻位诱导DNA组装技术,实现高效的靶标识别与信号转化,建立一种简单快速、性能稳定、成本低廉,可广泛用于复杂实际样本中低丰度核酸和蛋白质高灵敏、高特异性检测的通用型检测系统。并将该方法初步应用于前列腺癌患者血清PSA的检测,验证其临床诊断性能。本项目的实施将为生命科学研究和临床实验诊断提供有力的分析工具;同时为充分开发CRISPR/Cas系统的应用潜能提供新的思路和实验基础。

The sensitive detection of nucleic acids and proteins is of great significance for life science research and early diagnosis of diseases. Traditional detection techniques cannot meet the needs of scientific research or clinical laboratory because of their low sensitivity, complicated operation procedures and high cost. The CRISPR/Cas12a detection system has the advantages of simple, fast, sensitive and cheap, but it is difficult to be widely used in nucleic acid and protein detection due to the limitation of PAM recognition sites. On the basis of the previous work, this project intends to transform and optimize the CRISPR/Cas12a system by exploring the recognition mechanism of PAM locus to overcome the dependence of PAM sequence. With the combination of proximity-induced DNA assembly technology, efficient target recognition and signal transformation can be achieved, and a simple, fast, stable and low-cost universal detection system can be developed for the detection of low abundance nucleic acids and proteins in complex samples with high sensitivity and specificity. The method was applied to the detection of serum PSA in prostate cancer patients to verify its clinical diagnostic performance. The implementation of this project will provide a powerful analytical tool for life science research and clinical trial diagnosis. At the same time, it provides new ideas and experimental basis for fully developing the application potential of CRISPR/Cas system.
本项目实施以来,一直致力于建立基于DNA组装和CRISPR-Cas12a的通用型免预扩增高灵敏检测平台,构建了一系列简单实用、高灵敏、高特异的重要疾病标志物高效检测新方法。截至目前,已发表第一作者或通讯作者论文(含共同)SCI论文5篇,其中中科院一区4篇。主要研究内容包括:.(1)基于CRISPR-Cas12a的RNA定量检测方法的建立及应用. 利用T7外切酶介导的靶标RNA循环,将少量RNA靶标高效转化为大量DNA激活子,DNA激活子通过CRISPR/Cas12a的协同切割功能,切割连接的抑制链,释放检测信号。通过拆分G四链体结构组装的荧光信号变化产生可检测信号。该方法无需识别PAM序列,Cas12a克服对RNA靶标的限制,能够实现高灵敏、高特异性的RNA通用检测,并成功应用于临床尿液样本中的细菌16S rRNA分析。.(2)结合特异性分枝DNA阻断激活子与CRISPR-Cas12a方法建立FEN1检测策略. 通过DNA组装设计具有5′悬垂结构的特异性三通分枝DNA,利用其位阻效应阻碍BD激活Cas12a蛋白。通过引入基于荧光共振能量转移(FRET)的单链DNA报告探针,连续降解事件引起荧光信号强度的增强,实现了FEN1的高灵敏检测。.(3)基于CRISPR-Cas12a体系的单管等温检测平台用于TdT活性检测. 通过CRISPR-Cas12a协同切割和双重信号放大策略,实现了TdT活性的简单、高灵敏、高特异性检测,并对其他干扰蛋白表现出卓越的选择性。.(4)基于CRISPR-Cas12a协同切割的FEN1活性超灵敏检测. 建立了一种基于靶标诱导引物延伸以激活CRISPR-Cas12a协同切割的荧光生物传感器,用于超灵敏和特异性检测FEN1活性。该策略成功应用于血清和细胞裂解液中的FEN1检测,展示了潜在的临床应用前景。.(5)基于可控crRNA自转录和CRISPR-Cas12a策略用于FEN1活性检测. 开发了一种可控crRNA自转录辅助的双重放大CRISPR-Cas12a策略,用于高灵敏和高特异性检测细胞中结构选择性核酸内切酶FEN1。
