Improving the sugar content of pear fruit is a major goal in horticultural crop breeding. Nanhong is a high sugar accumulation bud sport of Nanguo. In our previous study, we measured the sugar content in Nanguo and Nanhong pear fruits and found that the sucrose content was higher in Nanhong. We compared the transcriptomes of the two varieties using RNA-seq and identified a sucrose transporter gene, PuSUT-like, as being expressed at higher levels in Nanhong fruit. To elucidate the PuSUT-like expression profiles in Nanguo and Nanhong fruits, we compared the PuSUT-like CDS from each; however, no difference was found. Moreover, no differences were observed in the promoter of PuSUT-like. A bHLH transcription factor PubHLH94 which was expressed at higher levels in Nanhong fruit could bind the promoter of PuSUT-like. However, the underlying mechanism of which sucrose levels can be elevated is still unclear. Based on these results, overexpression and silencing of PuSUT-like in pear fruit will be carried out to identify if the gene is essential for sucrose accumulation in pear fruit. Then, ChIP-PCR and GUS Reporter System experiments will be used to further clarify the transcriptional regulation model of PubHLH94 on PuSUT-like. Elucidating the PubHLH94 expression profiles in Nanguo and Nanhong pear fruits. Finally, overexpression and silencing of PubHLH94 in pear fruit will be carried out to clarify the gene function of how PubHLH94 regulating sucrose accumulation in pear fruit. Our research will be useful for understanding the mechanism underlying the sugar accumulation and providing evidence for creation of pear germplasm and regulation of fruit quality.
提升果实品质,尤其是提高其糖分含量一直是育种者的重要目标之一。本研究以南果梨及其高蔗糖积累芽变品种南红梨为试材,研究调控果实中蔗糖转运的分子机制。通过对两品种的果实进行转录组测序,发现与南果梨果实相比,PuMYB12和PuSUT4-like基因在南红梨果实中的表达水平较高,而PuPRE6和组蛋白去乙酰化酶PuHDAC9-like基因的表达水平较低。对PuSUT4-like蛋白进行亚细胞定位、在梨果实中进行基因沉默和过表达及酵母功能互补试验,发现PuSUT4-like蛋白定位于细胞膜,且对果实中蔗糖的运输至关重要。通过农杆菌介导的瞬时转化、酵母单杂交、ChIP-PCR及GUS试验,发现PuPRE6和PuMYB12拮抗调控PuSUT4-like基因的转录和蔗糖积累。PuMYB12通过正调控PuSUT4-like的表达促进蔗糖的积累,而PuPRE6通过抑制PuSUT4-like的表达抑进抑制蔗糖的积累。在果实中瞬时过表达PuPRE6和PuMYB12,发现PuPRE6抑制PuMYB12的表达,而PuMYB12不能调控PuPRE6的表达,通过酵母单杂交、ChIP-PCR及GUS试验,进一步验证了PuPRE6负调控PuMYB12的表达,上述结果表明PuPRE6位于PuMYB12的上游。转录调控可以在基因表达水平影响其基因积累情况,而表观遗传则是在蛋白水平调控蛋白的表达量。通过进一步研究发现南果梨果实中PuMYB12和PuSUT4-like基因启动子的组蛋白去乙酰化水平高于南红梨果实,且酵母单杂交实验表明PuHDAC9-like蛋白直接与PuMYB12和PuSUT4-like基因的启动子结合。我们的研究结果表明南红梨果实比南果梨果实呈现出更高的PuMYB12和PuSUT4-like基因表达量,以及更低的PuPRE6和组蛋白去乙酰化酶PuHDAC9-like基因表达量。PuPRE6通过与PuMYB12和PuSUT4-like基因的启动子结合来抑制它们的转录,而PuMYB12增强PuSUT4-like基因的表达。此外,在南红梨果实中,PuMYB12和PuSUT4-like基因的高表达水平与它们启动子和编码序列的高乙酰化水平相关。我们的研究结果揭示了梨中蔗糖积累背后新的转录调控和表观遗传机制。
