核与辐射安全与人群心血管健康的关系以往常被忽视,缺乏系统研究。目前,流行病学研究认为核与辐射会诱发心血管疾病增加,且我国核工业、核电及医疗等放射工作人群快速增长,辐射对该人群的健康影响不容小觑。核与辐射诱导心血管损伤基因、分子、靶点,以及低剂量辐射与缺血性心脏病的关系等重大科学问题尚待探索,应加以证实。ABCD4是溶酶体膜蛋白之一,维持细胞稳态及功能。我们首次发现低剂量辐射暴露人群ABCD4在不同剂量照射下显著低表达,且存在剂量依赖性,人群数据进一步显示辐射可增加心血管损伤风险。本项目将在此基础上,通过构建ABCD4过表达和基因沉默载体及实验动物模型,从分子、细胞、动物、人群等角度深入探讨ABCD4在辐射诱导心血管损伤中的作用及其机制。研究结果不仅可揭示ABCD4与辐射诱导心血管损伤的变化规律,还可为放射工作人员心血管损伤的防控和干预措施提供新线索、新思路。

It was formerly ignored and lacked of research that the relationship between radiation safety and cardiovascular health in the past. At present, it was believe that nuclear radiation may increase cardiovascular diseases from epidemiological studies, and with the rapidly growing of nuclear industry and nuclear power’ persons, it cannot be underestimated on human health effects from radiation. So it is extremely necessary to carry out relevant research. There major scientific issues, from radiation-induced cardiovascular damage related genes, molecules, targets, and the relationship between LDIR and ischemic heart disease, remain to be explored and confirmed. ABCD4, maintaining cell homeostasis and function, is one of the lysosomal membrane proteins. We found firstly that the expression of ABCD4 mRNA in the low-dose ionizing radiation exposure staffs was significantly reduced with a dose-dependent manner. The results of further preliminary experiments strongly prompt that ABCD4 is related to cardiovascular radiation damage. Base on this basis, this project will deeply explore the role and mechanism of ABCD4 in radiation-induced cardiovascular damage from multiple levels based on building ABCD4 overexpression and gene silencing vectors and experimental animal models. The research results can not only reveal the changing discipline of ABCD4 and radiation-induced cardiovascular injury, but also provide new clues and ideas for radiation workers' cardiovascular injury prevention and control and intervention measures.
心血管疾病已成为既肿瘤后的第二大死因,随着放射工作人员的逐年增加,由高脂饮食等诱发的放射性心血管疾病的健康问题不容忽视。本项目基于前期工作基础,开展ABCD4在放射性心肌损伤中的作用机制研究,并拓展了放射性动脉粥样硬化相关研究,包括电离辐射对心肌细胞的生物效应研究、放射性动脉粥样硬化实验动物模型验证、外周血经辐照后的动脉粥样硬化相关变化基因的生物信息学分析。不同剂量照射小鼠心肌细胞后发现低剂量照射ABCD4表达量下降不明显,而相对高剂量照射会增加ABCD4的表达。然而,在低剂量分割照射下,ABCD4的表达量变化不大,可见小鼠心肌细胞对低剂量辐射敏感性不大。不同剂量照射大鼠心肌细胞,发现4Gy以上会显著改变ABCD4等分子的表达量,siRNA敲低实验也发现大鼠心肌细胞经一定量的辐射后,可能会发生细胞凋亡、铁死亡等生物过程。动物实验发现,与未照射组比较,照射组中性粒细胞等白细胞会显著下降,高脂饮食组NETs 浓度明显升高,而辐照组在电离辐射的前两个月NETs 浓度基本呈现降低趋势,但差异并不显著。然后使用油红O染色小鼠主动脉,饱和油红O 染色结果与上述结果相对应,高脂饮食组相比对照组血管内壁斑块明显增多,而高脂+IR 组较单纯高脂组血管内壁斑块则显减少。生物信息学分析揭示了照射后和动脉粥样硬化数据集中的共享基因,表明免疫系统中性粒细胞可能是连接辐射和动脉粥样硬化的关键节点,中性粒细胞死亡释放的NETs会影响动脉粥样硬化的发展。认为极低剂量的照射可能不会额外增加放射性心血管损伤的风险,但长期低剂量辐射接触,尤其是长期高胆固醇饮食人群,应加强对放射性心血管健康风险的关注,保障该群体的健康。
