近几十年来人类的精液质量在不断下降,环境污染可能是导致其下降的主要原因,因为男性更容易受到环境污染物的影响。环境污染物可以破坏睾丸细胞间连接特别是血睾屏障从而导致精子发生异常。因此研究精子发生中细胞间连接是如何形成、重建,以及调控这个过程的主要因素对于我们研究环境污染导致的精子发生异常是非常有意义的。虽然大量研究发现乙酰胆碱M受体存在于动物睾丸细胞中并且可能参与精子发生,但还没有定论,而且还没有系统研究乙酰胆碱M受体在精子发生中的作用及机理。本申请拟在我们课题组已经发现的乙酰胆碱M5受体(M5R)通过影响细胞间连接来调控精子发生的基础上,来探究M5R在调控精子发生中的作用机制。主要是通过睾丸特异性地抑制M5R的表达来探究M5R在调控血睾屏障及各类细胞间连接的降解-重建中的作用,然后通过睾丸特异性的敲除M5R来验证其调控机理,以期为治疗男性生育力下降(少精/无精症)等难题提供理论依据。

In the past few decades, the quality of human semen (sperm motility and concentration) has been decreased continually. The environmental contamination may be the major cause for the human semen quality declining because males are more susceptible to the contamination. Moreover, the environmental contamination disrupts testicular cell-cell junctions especially blood-testis barrier (BTB) to lead to the abnormal spermatogenesis. Therefore, the investigation of the degradation-reconstruction of BTB and different cell-cell junctions and the regulatory factors is crucial to understand spermatogenesis. Although many studies have found that muscarinic acetylcholine M receptors are expressed in the mammalian testes and might be involved in spermatogenesis, the roles of them in spermatogenesis are not fully understood. Moreover, the underlying mechanisms of M receptors in spermatogenesis are not systemically investigated yet. Our preliminary study has found that muscarinic acetylcholine receptor M5 (M5R) regulated spermatogenesis through the modification of testicular cell-cell junctions. This investigation will explore the underlying regulatory mechanisms of M5R in spermatogenesis. The mouse testicular M5R will be specifically knocked down by shRNA and the role of M5R in the degradation-reconstruction of BTB and different cell-cell junctions will be deeply investigated. Then the testicular M5R will be conditionally knocked out to verify its role in the regulation of the degradation-reconstruction of BTB and different cell-cell junctions. The result from this investigation will provide fundamental knowledge for spermatogenesis, and a theoretical guide to improve the decrease in male fertility (oligozoospermia/azoospermia).
男性不孕不育是一个新兴疾病,这一疾病是继癌症和心血管病后的又一顽疾。世界范围内约有15% 的育龄夫妇不能生育,在这些不育男性人群里大约30%的因素是由于精子发生受阻从而导致非梗阻性无精症。近20年来精子发生这一科学前沿问题得到了广泛的关注,并得到飞速发展,这个问题是世界研究的热点、难点,因为精子发生是一个十分复杂、高度细胞内调控的细胞活动。精原干细胞要通过迁移从基质室到近腔室,要经历多次迁移最后形成精子。在这多次迁移中,支持细胞间的血睾屏障及各种细胞间连接要经历降解-重建的多个循环过程,因此在精子发生的过程中细胞-细胞间的各种连接对于精子的发生至关重要。然而细胞-细胞间连接的降解-重建是同时地、有序地、快速地进行着,是什么因素调控这种降解-重建现在还不清楚。乙酰胆碱M5受体可能参与睾丸支持细胞和生精细胞的增殖、支持细胞的分泌,但是它是否参与精子发生过程中生精细胞的迁移、及伴随迁移的细胞-细胞之间的连接来调控精子的发生还未知。本项目借助shRNA抑制睾丸乙酰胆碱M5受体(M5R),及Cre-loxP系统成功构建雄性小鼠睾丸支持细胞(AMH-cre启动)乙酰胆碱M5受体(M5R)特异性敲除鼠。发现睾丸支持细胞特异敲除M5R,导致雄性动物精子活力下降、繁殖力下降(怀孕率和产子数减少),这些结果说明M5R在精子发生中的起到重要作用。研究以雄性Flox纯合子(M5Rflox/flox)小鼠作为对照组,睾丸支持细胞双等位敲除鼠(cM5R)作为实验组,从发育学角度,收集不同时间点1周龄、3周龄、5周龄、8周龄雄性小鼠样品,从表型结构到分子机制,深入挖掘M5R在雄性睾丸精子发生,精子功能中的主要生物学功能,以期为探索神经递质调控(乙酰胆碱)调控精子发生过程机制研究提供帮助。这是首项研究揭示了M5R的调控精子发生的作用,丰富了我们对M5R在精子发生中调控机制的认识,为今后雄性不孕不育的治疗提供一种新的策略和技术支撑。
