既往研究表明童年期虐待与青少年自伤行为相关,但两者的因果关系及有关生物学机制需进一步阐明。基于前期研究,我们假设LINGO3和POU3F1去甲基化可介导童年期虐待影响青少年自伤行为易感性。本研究拟首先利用前期收集的样本验证外周血淋巴细胞中LINGO3和POU3F1去甲基化与青少年自伤行为的关联;然后利用课题组建立的青少年队列分析童年期虐待与唾液中LINGO3和POU3F1去甲基化的相关性;随访队列研究对象三年,推断童年期虐待与青少年自伤行为的因果关系并分析LINGO3和POU3F1去甲基化的中介作用。同时,运用动物实验探讨童年期不同照顾水平和母婴分离暴露对青春期大鼠情绪行为、海马和额叶皮层神经细胞生长及LINGO3和POU3F1去甲基化的影响。研究结果将可明确童年期虐待和青少年自伤行为的因果关系及LINGO3和POU3F1去甲基化的中介作用,为预防、诊断和治疗青少年自伤行为提供科学依据。

Previous studies suggested that childhood abuse was associated with non-suicidal self-injury susceptibility among adolescents, however the causal association of childhood abuse with non-suicidal self-injury and the related biological mechanism need to be further studied. Based on our previous studies, we hypothesized that demethylation of LINGO3 and POU3F1 could mediate the impact of childhood abuse on non-suicidal self-injury susceptibility. In this project, we will firstly use pre-collected samples to verify the association of LINGO3 and POU3F1 demethylation in peripheral blood lymphocytes with non-suicidal self-injury, then we will use the baseline of our adolescent cohort to analyze the association between childhood abuse and demethylation of LINGO3 and POU3F1 in saliva, and will follow up the cohort participants for three years to infer the causal association of childhood abuse with non-suicidal self-injury among adolescents, and to investigate the mediated effect of LONGO3 and POU3F1 demethylation. We will also use animal experiments to investigate the impact of different childhood care levels or mother-infant separation exposure on adolescent rats' emotional behaviors, hippocampal and frontal cortical nerve cell growth, and demethylation of LINGO3 and POU3F1. Our findings will clarify the association of childhood abuse with non-suicidal self-injury among adolescents, and the mediated effects of LINGO3 and POU3F1 demethylation, which could provide scientific evidence for the prevention, diagnosis and treatment of non-suicidal self-injury among adolescents.
非自杀性自伤行为(Non-suicidal self-injury,NSSI)严重影响青少年身心健康和发展,是青少年面临的重要精神卫生和公共卫生问题之一。近年有多个研究探讨了童年期虐待与NSSI的相关性,但二者的因果关系仍需循证,尤其缺乏生物学证据。结合课题组前期的研究基础,我们推断童年期虐待可能会改变某些基因甲基化水平的表达从而改变人群NSSI的发生风险。在本项目中,我们利用本课题组建立的中国青少年人群队列分析了童年期虐待、DNA甲基化、及青少年NSSI三者之间的关系,并结合了动物实验和细胞实验对上述在人群研究中得出的研究结果进行了验证。本研究发现的主要结果有:(1)包括童年期虐待在内的多种不良经历与青少年NSSI等多种身心健康问题存在关联;(2)并非所有的童年期虐待类型均与青少年NSSI显著相关;(3)采用不同方式评价童年期虐待对判断其与身心健康的相关性影响并不大。(3)童年期虐待能使多个基因甲基化水平发生变化,并且不同虐待类型和持续时间的童年期虐待对甲基化影响并不全相同;(4)童年期虐待影响青少年NSSI的易感性可能部分通过cg04622888、cg05037505甲基化中介,且上述两个甲基化位点仅中介情感虐待/忽视及躯体虐待/忽视与青少年NSSI之间的关联。(5)动物和细胞实验显示童年期虐待也能改变基因甲基化水平,其中甲基化水平发生变化的基因主要涉及激素生物合成过程、小脑发育等255个生物过程,涉及肌动蛋白丝结合、氧化还原酶活性等37个分子功能,以及涉及过氧物酶体等32个细胞成分。本研究深入阐述了童年期虐待与青少年NSSI之间的因果关系,不仅为相关疾病的精准防控提供可续依据,也为疾病诊断和治疗提供了新的靶点。
