深海冷泉、热液和深渊环境均属深海极端环境,是地球科学和生命科学的前沿研究领域。受地质活动和流体活动的影响,上述深海环境均处于动态变化中,是研究微生物环境适应性演变的理性对象。为系统揭示深海环境变化对微生物的短期和长期影响,本研究拟选择南海海马冷泉区、西北印度洋卡尔斯伯格脊卧蚕热液区和马里亚纳海沟等典型深海极端环境,利用沉积物岩芯样品和时间序列采集的近底水体样品,开展地球化学和微生物多组学的配套分析,研究昼夜-年际-千年际等多时间尺度下微生物种群、代谢功能及活性的演变特征;结合实验室模拟研究,揭示驱动深海微生物演变的主要环境因子及微生物与深海环境的互作机制;通过群体遗传学和系统发生学分析,研究代表性微生物类群对不同深海极端环境的适应策略及演化规律。本项目的开展将有助于系统认识深海环境微生物在不同时空尺度下的演替、适应和演化特征与过程,加深对微生物与深海环境相互作用、协同演化的认知。

The three typical deep-sea extreme environments, deep-sea hydrothermal vents, cold-seeps and hadal zones, are the most concerned research fields in both earth sciences and life sciences. The three typical deep-sea environments are constantly variable due to geological and fluid activities, providing ideal materials for the studies of microbial adaptation and evolution. This study aims to elucidate the short-term and long-term impacts of dynamic deep-sea physicochemical environments on microorganisms, as well as the evolutionary and adaptive characteristics of deep-sea microorganisms in response to environmental changes. By using sediment cored samples and time-serial bottom sea water samples collected from the cold-seep ‘Haima’, the Wocan hydrothermal plume-influenced zone in the northwestern Indian Ocean and the Mariana Trench, the succession of microbial community structure, metabolic functions and activities at different temporal and spatial scales such as diurnal, interannual, and millennial scales will be investigated by means of microbial omics and geochemical analysis techniques. By integrating analysis of environmental physicochemical parameters and simulation experiments, the study aims to reveal the primary environmental factors driving the evolution of deep-sea microorganisms and elucidate the mechanisms of microbial interactions with the deep-sea environment. Through population genetics and phylogenetic analysis, the study will investigate the adaptive strategies and evolutionary patterns of representative microbial groups in different deep-sea environments. This research is expected to provide a systematic understanding of the succession, adaptation, and evolution processes of microorganisms in typical deep-sea environments at different temporal and spatial scales, thereby deepening our knowledge of the interactions and coevolution between microorganisms and the deep-sea environment.
微生物在驱动深海碳氮硫等重要元素的地球化学循环中扮演着重要角色。然而,对于地质活动引发的环境变化如何影响深海生态系统,面对深海极端环境的选择压力,微生物经历了怎样的演变过程,目前的认识还非常有限。.本研究解析了深海热液、冷泉和深渊三种典型深海生境的微生物种群和代谢功能的时空演替规律,探讨了驱动微生物演替的主要环境因子及其作用机制。研究结果表明,水深是影响克马德克海沟微生物空间分布的关键因素,微生物种群结构在约6500米处出现明显变化。同时,在底部洋流和重力流等作用下,深渊微生物种群存在频繁波动。我们追踪了新生冷泉微生物的逐年变化。甲烷代谢类群的相对丰度变化提示,新生冷泉发育过程中甲烷代谢由有氧氧化转为厌氧氧化过程,微生物群落组装主要受扩散限制影响,同质选择的影响逐年降低,同时,微生物间相互作用亦逐渐减弱。对深海热液生态系统的研究表明,地震引发热液羽流的浊度异常升高,进而导致羽流区中热液来源古菌相对丰度的增加。此外,我们首次发现红外光对热液微生物种群结构的影响。深海热液细菌可以利用红外光抵御高压对细胞壁合成的抑制作用,从而适应深海高压极端环境。.在深海微生物适应性演化过程方面,本研究针对深海关键微生物类群开展了群体遗传和系统发生学分析,解析了主要海洋微生物类群的演化历史。我们发现Sulfitobacter的基因组系统发生关系与其物种演化关系存在明显差异,由此首次在原核生物中发现由不完全谱系分选机制驱动的快速物种分化。全球尺度下的系统发生学分析揭示了海洋主要微生物类群的5种海-陆演化模式及其代谢偏好性,发现了异化硫代谢微生物的快速分化与大氧化事件等重要的地质历史事件密切相关。.综上所述,本研究解析了三种深海典型生境中微生物适应环境变化的演替规律和机制,提出原核生物物种分化的新机制,重新理解了海洋微生物类群的演化驱动力,为深入认识微生物与深海环境相互作用、协同演化过程提供了关键信息。
