The complex nature of cell plasma membranes raised intensive research in the last decades, stimulated by the hypothesis of lipid microdomains. Cellular membranes are laterally heterogeneous and consist of transient and dynamic domains with varying properties, which prominently include liquid-disordered and liquid-ordered domains. Membrane domains can be induced and regulated by a variety of interactions to initiates a range of central biological and pathological events, which include specific lipid–lipid and lipid–protein interactions, and interactions between membrane components and the underlying cytoskeleton. The rearrangement of lipid microdomains can regulate cellular deformation, signal transduction and transmembrane transport processes through modulation of membrane tension,association and dissociation of membrane proteins. N,N’‑disubstituted- dihydrodibenzo[a,c]phenazines is a novel ratiometric dual-emission fluorescence probes based on vibration-induced-emission (VIE) mechanism, which is highly sensitive to changes in polarity and viscosity, and is an ideal probe for studying the dynamic processes of association between membrane proteins with lipid microdomains. This project employs a series of dual-emission unimolecular probe to visualize the dynamics change of lipid microdomains, aims to unravel the basis of intrinsic interaction between K-Ras protein and lipid microdomains, and provides fundamental insights into the physiology of cell membrane stress change, cell directional-migration, and inhibition of cancer cell spreading.
细胞膜微区的流动性改变和重排与包括细胞增殖、迁移、病原微生物感染等许多生理与病理过程密切相关,例如,细胞膜表面的配体-受体识别可能引发局部脂质重排和膜张力变化,从而介导内吞过程或激活特定信号通路,最终影响细胞命运。因而,对于细胞膜微区流动性受不同内源和外源性刺激而动态改变的原位探测将有利于膜生物学规律揭示。申请人利用振动诱导发光(VIE)这一独特的荧光机制,设计合成了一系列具有VIE发光机制的探针分子,选择不同磷脂组成及不同尺寸的脂质体(LMV和GMV)作为细胞膜模型,进行了磷脂膜微观结构的可视化研究。利用比例荧光示踪技术研究囊泡相变过程,并证明该探针可区分不同的膜微区并进行成像。此外,该探针成功示踪了stxb识别诱导的囊泡膜内陷过程,以及胆固醇浓度依赖的细胞膜微区变化,为细胞膜动态研究提供了新的研究方法。项目资助期间,以第一/通讯作者发表致谢SCI论文12篇,其中Chem(1篇), Angew. Chem. Int. Ed. (1篇)、Coord. Chem. Rev(2篇)。
