社会应激对人和动物的行为和神经内分泌系统会带来强烈影响,而目前社会应激的神经内分泌机制仅在雄性开展了深入研究,由于社会应激模型存在性别差异,所以孕期发生社会应激对母性行为的影响以及产后抑郁之间的关系还不清楚。五羟色胺是介导母性行为的重要神经递质之一,同时发现焦虑抑郁患者五羟色胺也存在异常。本研究拟利用高社会性的棕色田鼠为研究对象,建立孕期社会应激模型,通过母性行为的行为特征分析,再通过双重荧光免疫组化、实时定量PCR、western blot、选择性化学遗传学操控等多种手段从不同层面,阐述五羟色胺及其受体在社会应激的母性行为及产后情绪的影响及调节机制。通过该研究有望回答有关社会应激影响母性行为的生理生态学科学问题,还可以改进动物管理策略、减少社会应激、提高动物福利提供重要的理论依据,并可为保护成年女性产后精神健康提供新思路。

Social defeat occurs when an individual loses in an aggressive encounter, which robustly induces behavioral phenotypes related to anxiety and depression in naturalistic states. Almost all neuroscience studies using social defeat stress have used male rodents because adapting this protocol for females is challenging. It has been known that depression and anxiety are more prevalent in women than men for over 2 decades. The previous iteration of this grant determines that serotonin 5-HT2A and 5-HT2C receptors play distinctive roles in the regulation of rat maternal behavior. Activation of 5-HT2A or 5-HT2C receptors disrupts various maternal responses (e.g. pup retrieval, pup licking, nest building), while blockade of these receptors has no effect. Behaviorally, 5-HT2A and 5-HT2C affect maternal behavior through different behavioral mechanisms. Activation of 5-HT2C suppresses the motivated responses towards pups, while activation of 5-HT2A has little effect on maternal motivation. However, little is known about effect of social defeat on maternal behavior and interaction between social defeat in pregnancy and post-partum depression. The neural basis of 5-HT2C’s maternal effect, which is likely to involve the VTA projecting dopamine (DA) neurons and other non-DA neurons that express 5-HT2C.Here we establish Mandarin voles with higher sociability, suffering vicarious social defeat stress in pregnancy as model to examine maternal behavior, to analyze of maternal affect (e.g., pup preference), maternal motivation (e.g., pup retrieval on an elevated plus maze [EPM]), then to dissect the precise 5-HT2A and 5-HT2C functional neurocircuits underlying maternal behavior and post-partum depression with behavioral technology, double fluorescence immunohistochemistry, RT-PCR, Western-blot and designer receptors exclusively activated by designer drugs(DREADDs) technology. This study will enhance our understanding of the serotonergic neural mechanisms underlying maternal behavior and has implication for postpartum depression, and also provide a new insight to protect the health and well-being of mothers of human and wild animals.
孕期应激是产后各种精神疾患重要的诱因之一,对母亲和后代都会造成深远影响。本研究以孕期应激作为围产期压力来源,研究了孕期应激对母性行为和产后情绪的影响,并研究了对五羟色胺(5-HT)和相应脑区炎性因子的影响,进一步研究了多不饱和脂肪酸对孕期应激对母性行为和产后情绪的作用,探讨5-HT系统对孕期应激对母性行为和产后情绪影响的可能机制。.研究发现孕期应激会对母鼠的育幼行为产生影响。应激使母鼠体重显著降低,并会减少离开窝内幼仔的时间,这是母性焦虑的一种体现。在高架十字迷宫开臂末端嗅闻幼仔的次数明显增多。高架十字迷宫上幼仔暴露使应激母鼠中缝背核和正中缝核的表达减少,外侧僵核的c-fos表达增加。这些结果说明,孕期应激干扰了母鼠的育幼行为,可能使母鼠产生一种与资源限制相关的压力相似的过度警惕养育方式。应激导致母鼠产后行为异常可能与这些核团有密切联系,外侧僵核通过投射到中缝背核和正中缝核从而介导了孕期应激对母性行为的影响。.研究发现孕期补充多不饱和脂肪酸对母亲行为和异常情绪有改善作用。应激母鼠出现舔舐幼仔次数减少、高架十字迷宫上衔回幼仔的潜伏期延长,出现粗暴拖拽踩踏幼仔等异常育幼行为,并伴有产后有焦虑情绪,同时外侧僵核的5-HT含量减少,前额叶皮层和杏仁核5-HT1A受体减少,前额叶皮层炎性因子IL-1β、TNF-α、IL-6相对含量和MDA含量减少,而前额叶皮层和杏仁核cAMP含量增多,孕期对应激母鼠给予Ω-3脂肪酸后所有指标得以改善。说明-3脂肪酸对孕期应激损伤的母性行为改善的神经机制可能是通过5-HT1A型受体改变cAMP信号通路而从实现的。.本研究阐明了孕期应激使母鼠过度警惕养育方式,对产后焦虑抑郁的影响和5-HT系统之间的关系,以及多不饱和脂肪酸对其调节机制,为育幼行为和寻找产后焦虑抑郁提供新的治疗重要的理论依据和思路。
