EpCAM在恶性肿瘤进展中具有重要调控作用,在肿瘤诊断及靶向药物开发等方面受到诸多关注。然而,EpCAM复杂的调控机制及未知的生物学功能需进一步阐明。蛋白质的相互作用组学研究是了解目标分子新功能以及空间动态修饰的可靠手段。因此,本项目基于前期SILAC-AP/MS鉴定了EpCAM互作分子网络,并结合细胞器分级提取及蛋白定位的成像数据,探索EpCAM的亚细胞定位及进入细胞核的穿梭机制及功能意义。本项目将首次揭示细胞膜蛋白EpCAM转运入细胞核的现象,重点阐明EpCAM细胞核移位的运输机制及转运入细胞核的EpCAM的全新生物学功能。通过本项目研究,将有望揭示EpCAM新的亚细胞定位、空间动态调节以及全新的生物学功能,丰富EpCAM的理论基础研究,为基于EpCAM分子功能的肿瘤诊断及靶向治疗提供新的思路。

EpCAM is reckoned as a multi-functional tumor antigen involved in the regulation of malignant behavior of carcinoma cells, receiving considerable attention as a prognostic marker and a therapeutic target. However, there is unsolved discrepancy in the regulatory role and the prognostic value of EpCAM, which is remained unclear and requires further investigation. Herein we identified by SILAC-AP/MS a high-confidence EpCAM interactome consisting 77 novel interactors. We aim to integrate fractionated organelles analysis, protein-protein interaction network analysis and imaging of protein localization to uncover EpCAM subcellular localization and novel functions. Based on EpCAM interactome and previous investigation, we propose that the cell membrane-embedded EpCAM, through internalization utilizing endosome transport system as vehicle, transportin-1 as driver, translocates to the nucleus, where it binds to CCR4-NOT complex subunit CNOT1, leading to SLUG change through transcriptional reprogramming. This study focuses on 3 parts: The novel subcellular localization of EpCAM in endosome transport system and nucleus; The mechanism for nuclear entry of EpCAM; And the functional consequences of EpCAM nuclear translocation. This study will describe a novel function for EpCAM in transcriptional modulation through nuclear translocation of the internalized EpCAM and complex formation with CCR4-NOT transcription modulators. Given the importance of EpCAM in tumor development, this work will contribute to our understanding of molecular mechanism of EpCAM in cancer, and provide comprehensive insight into the future application in EpCAM-based translational medicine.
上皮细胞粘附分子(EpCAM)在上皮源性肿瘤细胞及胚胎干细胞中高表达,在调控循环肿瘤细胞、上皮间质转化、肿瘤干细胞干性维持等恶性演进进程中发挥重要作用,因此在肿瘤诊断、指导预后、液体活检、靶向药物开发等方面受到诸多关注。EpCAM在胚胎分化,组织形态建成,细胞分化及肿瘤细胞上皮间质转化中具有时空动态表达的特征,且此动态表达在肿瘤恶性演进及胚胎干细胞分化中发挥重要调控作用。本项目基于SILAC-MS鉴定了EpCAM的蛋白互作分子网络。分析发现EpCAM的互作分子与内体运输及膜蛋白循环密切相关,并进行互作及功能的验证,并在不同细胞系中通过细胞器分级提取及蛋白定位的成像数据验证了EpCAM的内吞,内体运输及膜循环。EpCAM表达在中胚层分化及肿瘤细胞上皮间质转化过程中呈现出动态下调的特征,此动态下调过程与EpCAM内吞增强及膜循环抑制密切相关。因此,内吞及膜循环介导的EpCAM时空动态是胚胎干细胞分化及肿瘤细胞EMT分化进程中调控EpCAM蛋白表达水平的重要方式。本项目进一步解析了EpCAM经内体系统运输及分选,与特异性介导蛋白入核的分子Transportin-1结合,介导EpCAM移位到细胞核。进入细胞核内的EpCAM通过CCR4-NOT复合体负向调控SLUG的表达,且该调控功能依赖于CCR4-NOT复合体亚基CNOT1。此外,本项目探明EpCAM可定位于线粒体膜,并对维持线粒体稳态具有正向调控作用。本项目除了探索EpCAM的亚细胞器定位以及全新功能外,也对EpCAM表达的调控机制以及EpCAM的受控膜内蛋白水解机制进行了深入的探索。阐明肿瘤细胞中NTS通过PIRIN上调EpCAM表达,以及作为配体促进EpCAM受控膜内蛋白水解释放可溶性胞外域EpEX到肿瘤微环境的新机制。本项目为肿瘤细胞上皮间质转化中EpCAM的表达调控机制提供了新的视角。揭示了EpCAM新的亚细胞定位、空间动态调节及其在调控肿瘤进展中的新功能,丰富EpCAM的理论基础研究,为将来研究EpCAM新的亚细胞定位及调控肿瘤进展的新功能提供了有价值的数据和平台。为基于EpCAM分子功能的肿瘤诊断及靶向治疗提供了前期基础。
