中药注射剂引起类过敏反应的物质基础是中药注射剂质量控制的重要内容。细胞膜色谱技术已被广泛应用于中药物质基础研究,基于野生型肥大细胞或MrgprX2受体过表达细胞的传统细胞膜色谱模型能够有效分析中药注射剂中的类过敏组分,但均存在稳定性差、特异性低的缺点,降低了对类过敏组分辨识的准确性及高效性。本课题拟构建Avi-tag-MrgprX2融合蛋白过表达的HEK293细胞,合成生物素键合材料,通过Avi-tag的生物素化定向固定MrgprX2受体,制备Avi-tag-MrgprX2新型固定相,有效提高固定相稳定性及对类过敏组分识别的特异性。构建Avi-tag-MrgprX2细胞膜色谱模型,并与高效液相色谱质谱联用构成二维分析系统,对中药注射剂中的类过敏组分进行精准分析。本项目制备的固定相能够实现对类过敏组分的高精度、高准确度分析,对中药制剂安全性评价具有重要的理论意义及应用价值。

The substantial basis of anaphylactoid reactions caused by traditional Chinese medicine injections is important for quality control of traditional Chinese medicine injections. Cell membrane chromatography technology has been widely used in the study of substantial basis. Combined with liquid chromatography-mass spectrometry, CMC can effectively screen, analyze, and further identify the active components related to anaphylactoid reactions in traditional Chinese medicine injections. Previous commonly used cell membrane chromatography models based on wild-type mast cells or MrgprX2 receptor overexpressed cells can effectively analyze allergic components in traditional Chinese medicine injections, but they all have shortcomings of poor stability and specificity, thus reducing the accuracy and efficiency of receptor-targeted identification for allergic components. This project intends to construct HEK293 cells overexpressing Avi-tag-MrgprX2 fusion protein, synthesize biotin-bonded carrier material, and achieve oriented/site-specific immobilization of Avi-tag-MrgprX2 cell membrane biomaterials through Avi-tag biotinylation. Hopefully, the subsequently prepared Avi-tag-MrgprX2 immobilized new stationary phase can effectively enhance the stability of the stationary phase, improve the specificity of the chromatography model to allergic components. The Avi-tag-MrgprX2 cell membrane chromatography model is constructed and combined with high performance liquid chromatography mass spectrometry to form a two-dimensional analysis system for the purpose of achieving accurate analysis of allergic components in traditional Chinese medicine injections. The new stationary phase prepared in this project can realize high-precision and high-accuracy analysis for allergic components, also has important theoretical significance and application value for the safety evaluation of traditional Chinese medicine preparations.
中药注射剂引起类过敏反应的物质基础是中药注射剂质量控制的重要内容。类过敏反应指初次使用药物即在患者体内引发过敏样症状,病人发生类过敏反应会加重病情,严重者可出现呼吸系统衰竭,甚至危及生命。肥大细胞是类过敏反应的效应细胞,其表面高度表达的MrgprX2被证实是药物引发类过敏反应的直接靶标。因此,对中药注射剂中靶向MrgprX2的类过敏组分进行分析尤为重要。.细胞膜色谱技术(CMC)已被广泛应用于中药物质基础研究,基于短肽标签的新型CMC固定相可有效提高固定相的稳定性、特异性,降低非特异性吸附。本研究以MrgprX2为研究对象,利用Avi-tag探索一种通过体外生物素化制备CMSP的新方法,建立稳定性好、特异性强的CMC模型及MrgprX2/CMC-HPLC/MS二维联用系统,用于筛选中药注射剂中的类过敏组分,并寻找靶向MrgprX2的新型拮抗剂。.本项目成功构建了MrgprX2-Avi-tag HEK293高表达细胞株,验证了2-亚氨基生物素(IB)为BirA的底物,利用Avi-tag与IB在BirA催化下的特异性反应,将含有MrgprX2-Avi-tag的细胞膜碎片键合于IB修饰硅胶(SiO2-IB)表面,制得新型CMSP。采用扫描电子显微镜、透射电子显微镜、免疫荧光、傅里叶红外光谱、X射线光电子能谱等手段证明了CMSP的成功制备。所制备的CMC柱稳定性好、特异性强、选择性良好、非特异性吸附降低,柱寿命大于7天。构建了MrgprX2-Avi-tag/CMC-HPLC/MS二维联用系统,用盐酸青藤碱验证了该系统可用于筛选、分析、鉴定作用于MrgprX2的活性成分。筛选发现舒肝宁注射液中的潜在致敏组分为绿原酸类化合物及野黄芩苷,且致敏组分在25 µM浓度下均能引发肥大细胞脱颗粒。此外,从蔓荆子、盐肤木、紫草、桑白皮提取物中筛选出蔓荆子黄素、漆黄素、紫草素、乙酰紫草素、桑根酮C、桑根皮素,药理学验证结果显示筛选出的活性组分具有抗类过敏的活性。.本研究制备的基于Avi-tag的CMC方法有效降低了固定相的非特异性吸附,建立的模型能够高通量快速的筛选作用于MrgprX2的活性化合物,为舒肝宁注射液导致类过敏反应的物质基础提供了依据,也为MrgprX2介导的类过敏相关疾病的治疗药物开发提供了一种新方法。
