Qinghai-Tibetan Plateau has the largest alpine grassland ecosystem in the world, which is sensitive to climate and environment changes. Due to natural and anthropogenic disturbances, the degradation of alpine grassland ecosystem on Qinghai-Tibetan Plateau becomes a serious problem. And the main restoration practices are artificial revegetation of the degraded grasslands. It has been demonstrated that arbuscular mycorrhizal fungi (AMF) play important roles in improving soil environment, promoting plant nutrient uptake, and helping ecological restoration. However, investigations on the response of AMF grassland restoration in Qinghai-Tibetan Plateau remain unclear. This project focuses on natural grassland and artificial grassland with different establishment years, explores the dynamics of AMF community structure, biomass and activity in the process of grassland restoration. By the combination of traditional biological methods and modern molecular methods, the effects of different environmental factors on AMF biological traits are analyzed. Using Mantel test, structural equation modeling and multiple stepwise regression, the relationship between vegetation restoration and AMF biomass, community structure and activity are clarified. The results of this study are expected lay the foundation for the application of AMF in revegetation practices, and provide theoretical basis for the management of alpine grassland ecosystems and biodiversity conservation.
