Carbon (C), nitrogen (N), and sulfur (S) are the most important compositions of living organisms, with complex and diverse forms for function or storage, and their cycling influences and even determines the complexity of the Earth's environment and the diversity of biosphere. Hydrosphere microorganisms obtain energy and nutrients to sustain their life activities, and this process produces and consumes organic matter, which mediates the change of C, N and S forms and drives the global biogeochemical cycle. Based on progress made during the last few years on the understanding of the mechanism of element cycles of C, N, and S, driven by microorganisms in the hydrosphere, the research teams involved in this project will focus on several research topics, which will aim to: reveal the molecular mechanism on the environmental sensing of the different elements, the regulation on bacterial metabolic network, and the feedback effect on food chain; to dissect the coordinated actions of microorganisms on the coupled assimilation of carbon/nitrogen at the coast wetland; to study the process and mechanism of C/N/S metabolism driven by °S in bacteria in coast water sediments; to understand the coupling mechanism of C/N/S metabolic balance, the new methane biosynthetic pathway in diazotrophic bacteria; to uncover new mechanism of microbial electron transfer powered by mineral thermal energy in deep sea, and the rules of mineral energy governing microbial metabolism; to elucidate the mechanism on extracellular-electron-driven C/N fixation and biofilm formation in microorganisms. Through the above research work at multiple levels from cell to community to representative ecosystems, we intend to reveal the new pathways and mechanisms of the hydrosphere microorganisms driving the Earth's elemental cycle, and explain the contribution of hydrosphere microorganisms as carbon and nitrogen sources and sinks, so as to serve the management and decision-making of the “dual carbon" strategy.
碳、氮和硫是生命体的主要组成元素,其存在形式复杂多样,并在循环过程中影响和决定着地球环境的复杂性及生物圈的多样性。本项目从微观和宏观层面揭示了水圈中关键元素参与生物地球化学循环的机制,并展示了相关研究的潜在应用价值。主要研究成果包括:揭示了蓝细菌中全局转录因子 PacR 和 DevH 在协调碳氮代谢方面的分子机制;发现了直接氨氧化微生物可先同化氨和有机氮为谷氨酰胺,再进一步氧化为羟胺和氮气的全新氮代谢途径;阐明了氮调控蛋白 PII 在转录和翻译后水平上对固氮酶转录激活因子 NifA 的调控机制;揭示了胞外氨肽酶通过调控群体感应信号分子来影响胞外多糖合成的新机制;发现了近海海底沉积物中零价硫驱动的细菌碳、氮、硫代谢过程及其机理;揭示了光合固氮菌与产甲烷古菌协同驱动甲烷生物合成的新机制,为光照在湿地甲烷排放中的影响提供了重要认识;通过实验模拟与理论计算,发现了水圈微生物协同矿物利用光、热等驱动碳、氮、硫元素循环的新途径与机制;开发了适用于深海等极端环境的压力下热力学数据库及定量预测工具,以研究矿物与微生物协同驱动元素循环的一般规律;揭示了滨海湿地淤泥质土壤硫循环紊乱导致土壤失衡的机制,并探讨了碱蓬-田菁间作等人工干预措施下的微生物协同作用;发现了草酸盐-碳酸盐转化可促进甲烷产生的微生物代谢新途径,并提出微生物诱导合成鸟粪石等生物矿物促进滨海湿地富营养化水体自净的原理,为滨海湿地淤泥质盐碱土及污染水体的生态修复提供了理论依据。本项目从细胞、群落及典型生态系统等不同层次,探究了水圈微生物驱动生物地球化学循环的新途径和机制,为揭示水圈微生物在碳氮收支中的作用及“双碳”战略的科学管理和决策提供了重要的理论支撑。
