英文摘要
Metaphase spindle formation is the fundament for the completion of cell division and correct segregation of chromosomes, while the centrosome as the microtubule-organizing center is required for the spindle formation in somatic cells. However, the role and mechanism of the centrosome in the formation of a meiotic spindle during spermatogenesis are unclear. We found that germ cell-specific protein TOPAZ1 was colocalized with centrosomal protein γ-Tubulin in mouse spermatocytes and its mutation did not affect the meiotic prophase I progression and crossover formation but resulted in the chromosomes not being correctly aligned on the equatorial plate. Further study showed that 85% of metaphase I spermatocytes of Topaz1 mutant mice displayed monopolar spindles. Therefore, we speculate that TOPAZ1 may promote the formation of bipolar spindles by regulating either the replication and separation of the centrosome or the ability of microtubule polymerization to ensure normal meiotic progression. This project will further clarify the effect of the Topaz1 mutation on centrosome behavior and spindle formation and start with the search for TOPAZ1 binding proteins, and then through a series of screening and functional verification, to find out new factors involved in the regulation of spindle assembly, not only to reveal the special regulatory mechanism of the formation of the meiotic bipolar spindle in spermatocytes but also to provide a theoretical basis for the diagnosis and therapeutic of infertility caused by related spermatogenesis disorders and genetic counseling.
精子和卵子由生殖细胞经减数分裂产生。在减数分裂前期I,同源染色体会发生联会和重组,通过形成交叉在彼此之间建立物理连接,以确保它们在中期I正确排列于赤道板和后期顺利分离。在雄性生殖细胞中,中心体作为微管组织中心对于纺锤体的形成至关重要。然而,雄性生殖细胞如何协调上述一系列复杂事件,至今尚不明确。在本项目研究过程中,我们发现TOPAZ1是一个特异调控雄性减数分裂纺锤体形成的蛋白。TOPAZ1呈液滴状聚集体定位于粗/双线期精母细胞中。Topaz1突变后,高达85%的中期I精母细胞纺锤体组装异常,表现为单极纺锤体。进一步研究发现,突变小鼠中心体虽能够正常复制,但其分离异常;同时,纺锤体极上中心体的PCM组分显著减少。通过转录组和蛋白组学分析,我们发现Topaz1突变后,一些中心体相关蛋白,如CDK5RAP2、WDR62和PLK1,其mRNA水平未发生显著变化,但蛋白含量显著降低,表明TOPAZ1可能是在翻译水平对这些蛋白的表达进行调控。有趣的是,借助酵母双杂交和免疫共沉淀联合质谱技术,我们发现TOPAZ1能够与GOLGA3、WDR62、EDC4和EIF3G等蛋白发生相互作用。基于此,我们认为TOPAZ1可能与EDC4和EIF3G形成复合物,以液液相分离的机制调控下游靶基因Plk1的蛋白表达量。PLK1可通过对WDR62进行磷酸化修饰,促进中心体的正确定位及纺锤体的组装;同时,PLK1还能对CDK5RAP2和PCNT进行磷酸化修饰,促进中心体PCM的成熟和稳定。后续,我们计划运用RIP-Seq技术和Ribo-Seq技术,进一步明确TOPAZ1与靶基因Plk1等之间的联系,从而验证上述猜想。受本项目支持,我们发现一种新的重组蛋白C12ORF40/REDIC1在减数分裂I类交叉重组中发挥着重要作用,相关研究成果于2023年发表在《Cell Discovery》杂志。此外,我们对哺乳动物联会复合体的结构、功能,及其突变与人类不孕不育的关联进行了全面总结与展望,该综述文章于2024年发表在《中国科学:生命科学》杂志。此外,申请人于2022年12月参加了一次国内学术会议,并于2023年6月参加了一次国际学术会议。在项目执行期间,还协助指导一名博士研究生完成博士毕业论文,并获得博士学位。
