The oligopeptide transporter 1 (PepT1) plays a crucial role in the transport and absorption of di-/tri-peptides in fish intestines. The preliminary studies have revealed that when the intestinal FoxOs activity of grass carp is activated or inhibited, the expression of CDX2 and PepT1 genes show significant synergistic changes. We speculate that FoxOs-CDX2 signaling pathway may have a regulatory effect on PepT1 transport di-/tri-peptides in the intestinal tract of grass carp. This project takes grass carps as the research object and uses cell biology, molecular biology and other technical methods to further identify the FoxOs-CDX2 signaling pathway in the intestinal of grass carp, and explore its regulatory effect on the expression of PepT1 gene. Then, we will clarify the regulation effect of grass carp transcription factor FoxOs on CDX2 promoter activity, as well as the response relationship and mechanism of grass carp FoxOs and CDX2 with intestinal PepT1 transit peptide. Eventually, the molecular mechanism of FoxOs-CDX2 signaling pathway regulating the intestinal PepT1 transport small peptides of grass carp was clarified, which provided theoretical basis and technical support for promoting the intestinal transport and absorption of di-/tri-peptides of fish and improving the feed protein utilization efficiency.
蛋白质是鱼类生长的关键营养素之一,蛋白质原料是鱼类配合饲料中最重要的组成部分。饲料蛋白在动物胃肠道消化的终端产物有超过一半以上是以小肽吸收进入体循环,供机体合成利用。其中,鱼类肠道上皮细胞小肽转运载体PepT1在动物肠道小肽的转运吸收中发挥着关键性作用。本项目以草鱼为研究对象,深入探究了FoxOs-CDX2信号通路调控草鱼肠道PepT1转运小肽的分子机制。首先,项目采用分子生物学技术,成功克隆FoxO3a、FoxO3b、FoxO4和CDX2等关键基因cDNA序列,利用生物信息学方法分析其分子特征和进化关系,鉴定了草鱼肠道FoxOs-CDX2信号通路;其次,采用荧光定量PCR、双荧光素酶报告基因系统、特异性激活或抑制表达等技术手段,揭示了FoxOs家族基因对CDX2基因启动子的调控作用,明确了FoxOs-CDX2信号通路对PepT1基因表达的调控机制;最后,通过营养学生长实验,探究白藜芦醇和丁酸钠等营养调控因子对草鱼肠道FoxOs、CDX2以及PepT1活性的影响,进一步验证了该信号通路在调节肠道PepT1转运小肽中的作用,从而系统阐明FoxOs-CDX2信号通路在草鱼肠道营养吸收中的重要作用。项目实施期间,团队成员共发表学术论文6篇,其中SCI收录4篇;申请国家发明专利2项;培养硕士研究生2名。研究成果不仅丰富了鱼类蛋白质营养吸收调控的理论体系,还为实际养殖中饲料配方优化和功能性饲料添加剂开发提供了重要的理论依据和技术支持。
