植物具有强大的再生能力以适应严苛的自然环境,这种能力已被广泛运用至农业生产,比如扦插、组织培养等。再生的本质是伤口或胁迫信号指导的细胞命运重编程,然而我们对其细胞学和分子机理的认识还十分欠缺。本项目前期的正向遗传学筛选表明受体激酶FERONIA参与根器官再生调控,其功能缺失突变体的根创始细胞无法正常分裂成根原基。经回复子筛选,我们定位到一个MAPKKK家族基因SOF225(SUPPRESSOR OF FERONIA 225)。初步研究表明再生过程中该基因高表达于根创始细胞,其低水平表达株表现出和fer相同的根再生缺陷,且在酵母中与FER的胞内域互作。我们推测其作为FER的下游调控根创始细胞的分裂以建立根原基。本项目将综合运用遗传学、生化及细胞学手段来揭示FER通过SOF225调控再生的细胞学和分子基础。这将帮助我们认识复杂而强大的再生现象背后的规律,同时在指导农业和园艺应用上具有重要意义。
英文摘要
Plants have evolved powerful regeneration ability to survive rigorous environment, which has been widely exploited in agricultural technologies such as cuttage and tissue culture for vegetative propagation. Regeneration is a process of the cell fate reprogramming upon wounding or stress. However the underlying mechanisms are poorly understood. Our previous genetic screen revealed that receptor kinase FERONIA was involved in root regeneration, its loss-of-function mutant could not establish root primordium from root founder cell division. Through suppressor screening, we identified a MAPKKK gene, SUPPRESSOR OF FERONIA 225 (SOF225). Preliminary studies showed SOF225 was highly expressed in root founder cells during root regeneration, and leaf explants from SOF225 knock down lines exhibited similar defects in root regeneration as in fer mutants. We propose that SOF225 may act downstream of FER to regulate root founder cells division to establish root primordium. In this project, genetics, biochemistry and cytology evidences would be provided to reveal the cellular and molecular basis of FER and SOF225 in root regeneration. This will help us to understand the powerful regulation pattern behind regeneration, and will also have important practical value in guiding the technology application in agriculture and horticulture.
植物能够通过器官再生的方式实现自我修复,甚至进行营养繁殖。植物再生的本质是在伤口或胁迫信号指导下的细胞命运重编程,这一过程依赖细胞命运转变和细胞分裂的协同进行,二者缺一不可。根再生研究领域已经揭示了一条清晰的细胞命运转变(生长素-WOX11-WOX5/LBD16)的调控线路,而根再生过程中细胞分裂是如何精确调控的却没有报道。在本项目中,我们发现受体激酶FERONIA(FER)参与拟南芥的根器官再生,其功能缺失突变体的根创始细胞无法正常分裂成根原基。通过正向遗传学方法筛选,我们发现MAPKKK7SN的突变可以回复fer根再生缺陷。利用这一回复基因,我们发现根再生过程中根创始细胞中的MAPKKK7定位于细胞质中,而在根原基中则定位于细胞核中。FER的胞内激酶域可以和MAPKKK7直接互作并对其磷酸化。我们的研究表明FER-MAPKKK7作为一个重要的功能模块调控根再生过程中根创始细胞的分裂从而建立根原基以启动不定根的发生。这些研究为植物器官再生领域提供了理论基础,也为将来将分子手段应用到再生技术提供了线索。
